RESUMEN

The mechanisms of anti-inflammatory action of saturated N-acylethanolamine--N-stearoylethanolamine (NSE) were investigated on the rat model of nonspecific inflammation (thermal burns of the skin). The results showed that the NSE application in a form of aqueous suspension (10 mg/ml) on the damaged skin area during 12 days significantly accelerated the healing process of burned wounds. NSE also prevented the increase of 11-hydroxycorticosteroids content in the blood of rats with burns. There was also found a significant decrease of cytokines (IL-1beta, IL-6 and TNFalpha) levels under the NSE action. This way may be one of the mechanisms of NSE anti-inflammatory action.

RESUMEN

The influence of N-stearoylethanolamine on the nitric oxide system, the state of lipid peroxidation, antioxidant enzyme activity, content of phospholipids and fatty acids were studied under the acute ethanol intoxication (2.5 g/kg) in rats. The results of investigations show that acute ethanol intoxication caused abnormalities of the oxidative homeostasis accompanied by the accumulation of thiobarbituric acid reactive substances (TBARS). High catalase and glutathione peroxidase activity was also shown. The altered content of total and individual fatty acids of phospholipids in the rat liver tissue was found. The content of saturated fatty acids (palmitic, stearic) increased and amount of unsaturated (palmitoleic, oleic, linolenic) acids decreased under acute ethanol intoxication. The changes of nitric oxide content was found in the brain, plasma and red blood cells. N-stearoylethanolamine (NSE) in a dose of 50 mg/kg of body weight shows the pronounced antioxidative and hepatoprotective properties under these conditions. It was found that the preliminary NSE administration to rats inhibited accumulation of TBARS and caused a simultaneous increase of antioxidant enzyme activity. The NSE administration modulated also the content of total and individual fatty acids of phospholipids and the amount of nitric oxide in pathologically altered tissues. These results suggested that NSE protected the structural integrity and functional ability of cell membranes under the acute ethanol intoxication.

RESUMEN

Effects of N-stearoylethanolamine (NSE), N-oleoilethanolamine (OEA) and mixture of more than 20 saturated and unsaturated N-acylethanolamines on phospholipids composition of normal and transformed fibroblasts in the culture were compared in the work. It was shown that cultivation of cells NSE decreases percent content of phosphatidylinositol (PI), phosphatidylserine (PS) and sphingomyeline (SM) - important mediators of cell signaling. Under the influence of OEA the level of SM decreases as well, but at the same time PI and PS levels increase. NAEs mixture decreases PS and PI levels in cells and causes phosphatidylcholine (PC) amount increase. Moreover NSE and NAEs mixture decrease the content of main mitochondria membrane lipid - diphosphatidylglicerole (DPG). OEA increases DPG amount. In transformed fibroblasts (line L929) NAE modulate lipid composition as well: NSE decreases the level of phosphatidylethanolamine (PE), OEA and NAEs mixture increase sphingomyeline levels. It is shown that response of normal and transformed fibroblasts on NAE application is different, depending on substance structure and cell-target type.

In experiments on isolated neonatal cardiomyocytes and hearts of adult rats we investigated the influence of polyunsaturated fatty acids (PUFA) on expression of two peroxisome proliferator-activated receptor (PPAR) target genes--fatty acid transport protein (FATP) and interleukin-1 receptor antagonist (IL-1ra). We determined that cell membranes of adult rats heart and cardiomyocytes of neonatal rats which were born by females fed with omega-3 PUFA for 4 weeks, contain more omega-3 PUFA (alpha-linolenic and docosahexaenoic acids) in comparison with the control. It was shown that omega-3 PUFA elevated FATP and IL-1ra mRNA expression in 2.25 (P = 0.07) and 8.4 (P < 0.00001) folds correspondingly in adult rats tissues. In isolated cardiomyocytes, the mRNA expression of FATP and IL-1ra were augmented 2.5 folds (P = 0.22) and 8.1 folds (P < 0.00001), accordingly. Our data show that omega-3 PUFA stimulate PPAR target genes expression and allow to permit genetic mechanisms in cardioprotective effects of omega-3 PUFA.

RESUMEN

The effect of N-stearoylethanolamine (NSE) on the lipid peroxidation process, antioxidant enzymes activity, phospholipid and fatty acid content in the rat liver tissues under acute morphine administration was studied. It was shown that morphine administration (30 mg/kg of body weight) caused an increase of the amount of thiobarbituric acid reactive substances (TBARS), alteration of antioxidant enzymes activity, decrease the protein level, quantity of total lipids and phospholipids, phosphatidylcholine, cholesterol esters; altered the content of some individual fatty acids. NSE administration (50 mg/kg of body weight) promoted normalization of the antioxidant enzymes activity and prevented the TBARS accumulation and decreased the total lipid and phospholipid quantity, increased the content of free and total cholesterol, corrected the level of free and individual fatty acids. It was assumed that NSE possessed antioxidative, membranoprotective and adaptive properties.

We investigated the influence of N-stearoylethanolamine (NSE) on tumor growth and metastasis of the lung Lewis carcinoma in mice. The effect of NSE on lipid composition of lung tissue under tumorogenesis was also studied. We demonstrated that NSE inhibited the tumor growth and decreased the volume and quantity of metastases being administered from the fourth day after injection of tumor cells to the last day of experiment and being administrated from the 21th day after injection of tumor cells to the last day of the experiment. The analysis of the lipid composition of the lung tissue showed the decrease of total phospholipid levels and change of the phospholipid spectra under tumor growth. The decreasing of the concentration of phosphatidylcholine, sphyngomyeline, phosphatidylserine and lysophosphatidylcholine in the lung tissue of tumor-bearding mice in comparison with lung of intact animals was observed. It was found that administration of NSE increased the level of lysophosphatidylcholine and decreased the concentration of phosphatidylinositol in investigated tissues. The content of sphingosine was increased in lung tissue of mice fed by NSE in comparison with tumor-bearing mice. The carcinoma development was associated by the significant decreasing of cholesterol level and by the increasing of unsaturated fatty acids in membrane phospholipids. The amount of the tiobarbituric acid (TBA) reactive substances in tumor-bearing mice was elevated. The administration of NSE inhibited the accumulation of TBA reacting compounds.

RESUMEN

Influence of NSE on lipid composition of metastases and the neighbouring conditionally normal lung tissue in mice with Lewis carcinoma was investigated. The processes of peroxidation in investigated tissues were also studied. It was shown that under the influence of NSE the high level of antioxidant activity in the metastases was decreased, while in the neighbouring conditionally normal lung tissue the catalase activity was increased. The content of the thiobarbituric acid-reactive substances in comparison with animals which were not fed by NSE was decreased. The development of carcinoma was accompanied by significant decrease of cholesterol level and by the increase of unsaturated fatty acids esterified in membrane phospholipids in both the metastases and the neighbouring conditionally normal lung tissue. An analysis of the phospholipid spectra shows that under tumor growth in investigated tissues the high-level lysophosphatidylcholine (LPC) was observed. The content of phosphatidyl choline (PC), phosphatidyl ethanolamine (PE), phosphatidyl serine (PS) was found to be significantly lower than in the lung of intact animals. It was found that administration of NSE to tumor-bearing mice contributed to the increase of cholesterol level, to the decrease of omega-6/omega-3 ratio polyunsaturated fatty acids of total phospholipids. NSE modulated the phospholipid membrane composition in both the metastases and the neighbouring conditionally normal lung tissue.

RESUMEN

The effect of N-acylethanolamines mixture (NAE) with saturated and unsaturated acyl chains on the fatty acid composition of the rat brain under chronic morphine dependence was investigated. Long-term administration of NAE reduced morphine-induced decrease of mono- and polyunsaturated fatty acids in the rat brain crude lipid extract. Furthermore, NAE restored the acyl chain spectrum, especially the content of docosahexaenoic acid in essential phospholipids--phosphatidylcholine and phosphatidylethanolamine. Pharmacological activity of NAE depended on a dose.

The influence of saturated and unsaturated N-acylethanolamines (NAEs) mixture on the rat brain lipid composition under the chronic morphine dependence was studied. It was shown that the long-term administration of NAE to rats with experimental morphine dependence restored the morphine-induced alterations of the brain phospholipid composition. This effect can probably be explained by a fatty acyl transfer from NAE to sn-1 position of some brain glycerophospholipids. Another reason of NAE beneficial effect could be due to its antioxidative property, thus providing the remodelling of phospholipid composition. The restoration of the brain essential phospholipids is associated with the decline in morphine consumption in rats with experimental morphine dependence. The mechanism of this phenomenon requires further investigations.

RESUMEN

The influence of irradiation and NSE on the microsomal lipid composition of the rat liver and heart was studied. It was shown, that radiation treatment in a dose of 2 Gy had not a significant affect on the heart phospholipid composition, whereas in the liver the amounts of the phosphatidylethnolamine and phosphatidylinositol were increased and the amounts of the phosphatidylcholine and sphyngomieline were decreased. NSE did not impact on these characteristics. The alterations of the plasmalogen/diacyl forms ratio of the PC and PE also took place only in the liver microsome. The analysis of the fatty acids esterified to phospholipids showed similar situation: the fatty acids of the heart microsome less were changed than those of the liver microsome undergo irradiation. The amount of arachidonic acid rise after the adding of NSE to rats. Investigation of the important component of biological membranes--cholesterol and its esters also showed that the liver tissue is more vulnerable to irradiation than the heart. NSE led to insignificant increase of the not esterified cholesterol in the liver. As a result of the present work we may conclude that the liver microsome is more vulnerable than the heart under X-ray radiation and NSE has protective effect in these conditions.

RESUMEN

Changes of the individual phospholipid fatty acid composition under the normothermal short-time ischemia with following reperfusion were investigated. Modification of the phospholipid fatty acid (FA) composition under ischemia-reperfusion didn't bear total character and was more manifested in cardiolipin (CL) and phosphatidylethanolamine (PE). The decrease of short chain FA in these phospholipids (more than by 50%) was observed. The amount of unsaturated FA included in CL increased and whole the saturated ones decreased. This caused the rise of the unsaturation index. The selective type of the changes suggested that they had an adaptive character. The addition of the N-stearoilethanolamine (NSE) into the perfusion solution caused a normalization of saturated and unsaturated FA relative amount, as well as of omega-3 and omega-9 FA level in CL. The modification of the FA composition of phosphatidylcholine (PC), phosphatidylserine (PS) and phosphatidylinositol (PI) was also found. The quantity of arachidonic acid in PC increased by 26% and the amount of stearinic acid enhanced in PS. The labeled N-([1-(14)C]-palmitoil)-ethanolamine was found in different lipid classes of the rat organs immediately 5 min following intraperitoneal injection. Approximately 1/3 of all incorporated label accumulated in the phospholipid fraction, and more than 50% of the labels were found in CL.

The effect N-stearoilethanolamines on lipide classes and their fatty acid composition was studied in the isolated rat heart under ischemia--reperfusion. It was shown that the amount of cholesterol esters increased at ischemia--reperfusion by 4.6 fold. The addition of 10(-7) M, 10(-6) M and 10(-5) M of N-stearoilethanolamines into perfused solution prevented these negative consequences of the myocardium ischemic injury. The addition of N-stearoilethanolamines decreased the quantity oleinic (18:1 omega 9) and free n-6 fatty acids. N-stearoilethanolamine lowered the amount of short chain saturated fatty acids and also polyunsaturated n-6 fatty acids in cholesterol esters. As well the lipide composition of perfused solutions was also investigated.

The effect of N-palmitoylethanolamine (NPE, 10(-5) M) on the lipid peroxidation (LPO) processes, phospholipid and fatty acid content in the rat liver at perfusion and ischemia during the liver preservation was estimated. As early as at the 5th min of perfusion by cooled conserving solution "Eurocollins", LPO activation was determined. Simultaneously the content of lysophosphatidylcholine (LPC) and phosphatidylserine increased, the total cholesterol level decreased. The redistribution of saturated and unsaturated fatty acids quantity was detected. The addition of NPE into "Eurocollins" solution reduced the accumulation of malonedialdehyde and LPC, modified the fatty acids content. These effects, evidently, formed the basis of the protective action of NPE on the hepatic tissues under anoxia.

The erythrocyte ghosts fatty acid composition of 27 patients with coronary artery disease (CAD) and 6 healthy people were investigated. Patients suffering from CAD were divided in two groups. The first group was matched by 18 patients with CAD and essential hypertension (EH) and 9 patients with CAD without EH. Significant decrease of the levels of some saturated fatty acids such as palmitic, stearic and begenic acids in the erythrocyte ghosts of the patients with CAD without EH was established among the major changes of the erythrocyte ghosts fatty acid composition. At the same time the amount, of arachidonic acid increased by 74% and docosahexaenoic acid--by 5.7 times. The content of miristic, palmitic and stearic acids was also diminished in patients with CAD and EH. The percent of arachidonic acid was elevated by 51% and docosatrienic acid--by 94%. It is notable that the quantity of docosahexaenoic acid did not change markedly if copmare to healthy people but was fallen for 5 times if compare to patients with CAD without EH. It was suggested that the lack of adaptive changes in erythrocyte ghosts omega-3 fatty acids of patients with CAD and EH was an important factor which could rise the risk of heart attack development.

The effect of 10(-5) M N-palmitoylethanolamine (NPE) on lipid peroxidation processes, phospholipid and fatty acid content in the ischemic rat liver under reperfusion was estimated. It was shown the significant accumulation of thiobarbituric acid reactive substances (TBARS), the increase of lysophosphatidylethanolamine (LPC), phosphatidylserine, phosphatidylethanolamine, sphingomyelin, general cholesterol content and also the decrease of phosphatidylcholine amount in the ischemic rat liver under reperfusion. Simultaneously the redistribution of saturated and unsaturated fatty acids quantity was detected, that indicates the increasing of membrane rigidity. The addition of NPE into the Eurocollins and Bretshneider's solutions under perfusion and preservation of a donor organ reduced the accumulation of TBARS, LPC, total cholesterol, prevented the changes of cholesterol/phospholipids and saturated/unsaturated fatty acids ratio value in the same experimental conditions. These effects, evidently, formed the basis of the protective action of NPE on the ischemic liver tissues under reperfusion.

RESUMEN

The brain phospholipid content was studied in rats with morphine physical dependence. It was found that level of total lipids, total phospholipids and cholesterol was diminished. The significant decrease of absolute amounts of phosphotidyl serine, phosphotidyl inositol, sphingomyelin and phosphatidyl choline was shown. At the same time the percent relation of individual phospholipids did not change. As the exclusion, the quantity of phosphatidyl ethanolamine plasmalogen lowered in rats with morphine dependence. The percent content of fatty acids 16:0, 18:1 omega 9, 20:1 omega 11, 20:4 omega 6 and 22:5 omega 3 was found to be reduced. At the same time the percent amount of i18:0 and 20:0 was shown to increase. As a result the unsaturation index dramatically diminished. It was also shown that under morphine physical dependence the acyl residues of phosphatidyl choline and phosphatidyl ethanolamine changed. It was concluded that lipids play significant role in the development of morphine physical dependence.

RESUMEN

The erythrocyte ghosts fatty acid composition of 29 patients with essential hypertension of the II stage were investigated. The significant decrease of some saturated fatty acids as myristic, palmitic and stearic under hypertension was established. Simultaneously the percent of omega-6 arachidonic and docosatrienoic acids substantially increased. At the same time no changes in omega-3 fatty acids were observed. The possible relationship between hypercholesterolemia and erythrocyte membrane fatty acid profile under essential hypertension discussed.

RESUMEN

The phospholipid composition of the heart of intact and experimental animals has been studied under conditions of simulated acute myocardium ischemia. It is shown that already at the 10th min the phospholipid composition of the heart ventricles changes expressively. The absolute amount of phosphatidylcholine, phosphatidylserine, diphosphatidylglycerol and phosphatidylglycerol decreases reliably. In this case lysophosphatidylethanolamine which is absent in control rats is identified. The amount of other phospholipids does not vary. Simultaneously the percentage of individual phospholipids varies: the phosphatidylethanolamine level significantly increases against the decrease of phosphatidylcholine and phosphatidylserine. As a result of the study of the amount of acylic and plasmalogenic forms of phospholipids it has been found that the level of the diacylic form of phosphatidylcholine in experimental animals falls. The data obtained testify to a definite participation of phospholipids in the pathogenesis of the acute myocardium ischemia.
