Ketosis metabolic research on lactating dairy cattle has been conducted worldwide; however, there have been very few Korean studies. Biofluids from lactating dairy cattle are necessary to study ketosis metabolic diseases. Six Holstein cows were divided into two groups (healthy (CON) and subclinical ketosis diagnosed (SCK)). Rumen fluid and milk samples were collected using a stomach tube and a pipeline milking system, respectively. Metabolites were determined using proton nuclear magnetic resonance (NMR) spectroscopy and they were identified and quantified using the Chenomx NMR Suite 8.4 software and Metaboanalyst 5.0. In the rumen fluid of the SCK group, butyrate, sucrose, 3-hydroxybutyrate, maltose, and valerate levels were significantly higher than in the CON group, which showed higher levels of N,N-dimethylformamide, acetate, glucose, and propionate were significantly higher. Milk from the SCK group showed higher levels of maleate, 3-hydroxybutyrate, acetoacetate, galactonate, and 3-hydroxykynurenine than that from the CON group, which showed higher levels of galactitol, 1,3-dihydroxyacetone, γ-glutamylphenylalanine, 5-aminolevulinate, acetate, and methylamine. Some metabolites are associated with ketosis diseases and the quality of rumen fluid and milk. This report will serve as a future reference guide for ketosis metabolomics studies in Korea.

Microbiota plays a critical role in the overall growth performance and health status of dairy cows, especially during their early life. Several studies have reported that fecal microbiome of neonatal calves is shifted by various factors such as diarrhea, antibiotic treatment, or environmental changes. Despite the importance of gut microbiome, a lack of knowledge regarding the composition and functions of microbiota impedes the development of new strategies for improving growth performance and disease resistance during the neonatal calf period. In this study, we utilized next-generation sequencing to monitor the time-dependent dynamics of the gut microbiota of dairy calves before weaning (1-8 weeks of age) and further investigated the microbiome changes caused by diarrhea. Metagenomic analysis revealed that continuous changes, including increasing gut microbiome diversity, occurred from 1 to 5 weeks of age. However, the composition and diversity of the fecal microbiome did not change after 6 weeks of age. The most prominent changes in the fecal microbiome composition caused by aging at family level were a decreased abundance of Bacteroidaceae and Enterobacteriaceae and an increased abundance of Prevotellaceae. Phylogenetic investigation of communities by reconstruction of unobserved states (PICRUSt) analysis indicated that the abundance of microbial genes associated with various metabolic pathways changed with aging. All calves with diarrhea symptoms showed drastic microbiome changes and about a week later returned to the microbiome of pre-diarrheal stage regardless of age. At phylum level, abundance of Bacteroidetes was decreased (p = 0.09) and that of Proteobacteria increased (p = 0.07) during diarrhea. PICRUSt analysis indicated that microbial metabolism-related genes, such as starch and sucrose metabolism, sphingolipid metabolism, alanine aspartate, and glutamate metabolism were significantly altered in diarrheal calves. Together, these results highlight the important implications of gut microbiota in gut metabolism and health status of neonatal dairy calves.

Korean peninsula weather is rapidly becoming subtropical due to global warming. In summer 2018, South Korea experienced the highest temperatures since the meteorological observations recorded in 1907. Heat stress has a negative effect on Holstein cows, the most popular breed of dairy cattle in South Korea, which is susceptible to heat. To examine physiological changes in dairy cows under heat stress conditions, we analyzed the profiles circulating microRNAs isolated from whole blood samples collected under heat stress and non-heat stress conditions using small RNA sequencing. We compared the expression profiles in lactating cows under heat stress and non-heat stress conditions to understand the regulation of biological processes in heat-stressed cows. Moreover, we measured several heat stress indicators, such as rectal temperature, milk yield, and average daily gain. All these assessments showed that pregnant cows were more susceptible to heat stress than non-pregnant cows. In addition, we found the differential expression of 11 miRNAs (bta-miR-19a, bta-miR-19b, bta-miR-30a-5p, and several from the bta-miR-2284 family) in both pregnant and non-pregnant cows under heat stress conditions. In target gene prediction and gene set enrichment analysis, these miRNAs were found to be associated with the cytoskeleton, cell junction, vasculogenesis, cell proliferation, ATP synthesis, oxidative stress, and immune responses involved in heat response. These miRNAs can be used as potential biomarkers for heat stress.

The microbial community within the rumen can be changed and shaped by heat stress. Accumulating data have suggested that different breeds of dairy cows have differential heat stress resistance; however, the underlying mechanism by which nonanimal factors contribute to heat stress are yet to be understood. This study is designed to determine changes in the rumen microbiome of Holstein and Jersey cows to normal and heat stress conditions. Under heat stress conditions, Holstein cows had a significantly higher respiration rate than Jersey cows. Heat stress increased the rectal temperature of Holstein but not Jersey cows. In the Kyoto encyclopedia of genes and genomes (KEGG) pathway analysis, Jersey cows had a significantly higher proportion of genes associated with energy metabolism in the normal condition than that with other treatments. Linear discriminant analysis effect size (LEfSe) results identified six taxa as distinguishing taxa between normal and heat stress conditions in Holstein cows; in Jersey cows, 29 such taxa were identified. Changes in the rumen bacterial taxa were more sensitive to heat stress in Jersey cows than in Holstein cows, suggesting that the rumen mechanism is different in both breeds in adapting to heat stress. Collectively, distinct changes in rumen bacterial taxa and functional gene abundance in Jersey cows may be associated with better adaptation ability to heat stress.

BACKGROUND: Although upper gastrointestinal (UGI) endoscopy is highly sensitive for the detection of esophago-gastroduodenal lesions, pain and discomfort during the procedure cause examinees to experience stress and anxiety. Moreover, there have been only a few studies on relief of pain and discomfort during UGI endoscopy through preparatory interventions. Therefore, the aim of this study was to investigate the relationship between a preparatory education program and the discomfort and retching experienced by examinees during endoscopy. METHODS: A total of 306 examinees who visited a health promotion center and underwent non-sedated endoscopy from May 13 to July 3, 2009 were included in this study. After they were assigned to experimental (n = 154) and control groups (n = 152), their discomfort and retching were measured with a visual analogue scale. The preparatory education program consisted of cognitive intervention, behavioral intervention and information. RESULTS: The preparatory education program relieved discomfort during endoscopy in male subjects, in subjects aged 60 and over, or in subjects with previous endoscopic experience with statistical significance (P < 0.05). It also relieved retching during endoscopy in subjects aged 60 and over with statistical significance (P = 0.023). Multiple logistic regression analysis showed that the preparatory education program significantly relieved the discomfort of examinees during endoscopy (P = 0.028). CONCLUSION: We found that the preparatory education program used in this study could significantly relieve the discomfort caused by endoscopy, particularly in subjects aged 60 and over, or in male subjects with a high incidence of stomach cancer in Korea.

BACKGROUND: For an early detection and prevention of dementia, there are growing concerns about the high-risk group for dementia and mild cognitive impairment. With an increase of obesity and its complications, obesity has become a major public health problem. Therefore, the aim of this study was to investigate the relationship between obesity and the high probability of dementia. METHODS: This study was done with subjects aged 60 to 89 years who visited a health promotion center, from April 1, 2008 to April 30, 2008. A total of 293 subjects (164 males and 129 females) were included in the evaluation of cognitive function using the Korean version of the Seven Minutes Screen test. Logistic regression models were used to analyze the association between obesity and the high-risk group for dementia. RESULTS: Among a total of 293 subjects, 71 subjects (29 males and 42 females) had a high probability of dementia. When compared to normal body mass index (BMI) group, overweight and obesity groups had about a 2.2-fold and 2.4-fold higher probability of dementia (95% confidence interval [CI], 0.97 to 4.99 and 95% CI, 1.07 to 5.46, respectively). When compared to non-obese and non-abdominal obesity group, non-obese and abdominal obesity group and obese and abdominal obesity group had about a 1.5-fold and 2.0-fold higher probability of dementia (95% CI, 0.59 to 3.95 and 95% CI, 1.09 to 3.84, respectively). CONCLUSION: This study shows that as the BMI increased, subjects had a higher probability of dementia; additionally, it suggests that abdominal obesity could be related to a higher probability of dementia.

BACKGROUND: The telemedicine services in Korea are expected to rapidly expand its use to the general population due to the development of digital networking, and its recent revision of related law and regulations. The purpose of this study was to investigate the knowledge and attitude of telemedicine in primary care patients. METHODS: We enrolled a total 243 participants, visited Family Medicine clinics and health promotion centers of university hospital in Seoul metrocity and Chungju city from April 1, 2010 to May 31, 2010. Data was collected by questionnaire, including demographic variables, knowledge and attitude of telemedicine. RESULTS: Among the total of 243 participants, 117 (49.8%) respondents were aware of telemedicine, and 178 (73.3%) respondents preferred telemedicine. The awareness showed differences according to the residence (P = 0.007), education (P = 0.03), and occupation (P = 0.02) of the respondents. The patient preference showed the differences at 50 years of age (P = 0.01) and in income of the participants (P = 0.005). CONCLUSION: Awareness of telemedicine in primary care patients was low. As for the patients more than 50 years of age who will be having difficulty manipulating the instruments, more education is crucial. Establishment of appropriate plans to increase patient preference is needed, especially for patients with low-incomes.

In this study, the free-radical-scavenging properties of anthocyanin oligomers for 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, alkyl radical, and hydroxyl radical were evaluated using electron spin resonance (ESR) spectroscopy. The DPPH radical, alkyl radical, and hydroxyl radical scavenging activity of anthocyanin oligomers increased in a dose-dependent manner, with the 50% inhibitory concentration (IC50) value of 13.0, 14.0, and 448.0 µg/mL, respectively. The inhibitory effect of anthocyanin oligomers on lipid peroxidation was examined with ferric thiocyanate (FTC) and thiobarbituric acid (TBA). The inhibitory activity of anthocyanin oligomers was found to be comparable to that of vitamin E. In addition, anthocyanin oligomers enhanced the activities of superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), glutathione peroxidase (GPx, EC 1.11.1.9), and glutathione-S-transferase (GST, EC 2.5.1.18) in ARPE-19 cells. In addition, anthocyanin oligomers inhibited the H2O2-induced G2/M phase arrest in ARPE-19 cells. Taken together, the present results demonstrate that anthocyanin oligomers have high antioxidative activity.
