RESUMEN

The genetic analysis of the Crimean-Congo hemorrhagic fever (CCHF) virus circulating in Stavropol region during 2011 year was suggested. A total of 14 RNA isolates from the Crimean hemorrhagic fever patients were genetically typed. The genetic analysis of the CCHF virus stains based on M-segment sequences (positions 2607-2932) supported the circulation of the genotype Europe 1 in the Stavropol region of Russia. In addition to previously known lineage STV-ROS, the second lineage VLG/ROS was observed in Stavropol region.

RESUMEN

Molecular epidemiological study of novel strain of Rubella virus isolated during the outbreak in Western Siberia in 2004 was described. Detailed phylogenetic analysis performed based upon entire SP-region, which encodes all three Rubella structural proteins (C, E2, and E1), was implemented. This analysis provides characterization of this strain and classifies it as 1H genotype, thereby correcting previous classification of this strain based upon shorter nucleotide sequence, only encoding E1 protein. Therefore, this study identified the genotype of the Rubella virus not previously detected in Western Siberia (and even entire Russian Federation), which highlights the importance of more extensive characterization of genetic variability of the Rubella virus, especially with regard to potential influence of vaccination on the Rubella virus mutagenesis.

RESUMEN

Evaluations of immune system of 155 patients with rubella and 90 contacts with patients were examined. Detection of viral genetic material in blood, urine, and nasopharyngeal swabs has been performed using RT-PCR method. Clinical diagnosis has been confirmed by RT-PCR in 114 (73.5%) patients. Changes of laboratory tests for rubella without clinical signs of the infection were observed in 20% of contacts. Complex ELISA- and PCR-assisted examination of patients can help to determine the stage of disease and characteristics of immune response. For differential diagnostic of rubella and other infectious diseases with exanthema it is rational to perform complex examination of patients using immunologic and molecular biologic methods.

RESUMEN

Two outbreaks of rubella infections notified in the Tomsk and Kemerovo Regions were investigated. Two rubella virus strains from one patient in each outbreak were isolated and genetically characterized. Reverse transcription polymerase chain reaction was used to reveal partial E1 gene sequence at a length of 915 nucleotides. Analysis indicated that the rubella virus strains circulating in the West-Siberian region belonged to international genetic 1g group, which had been first detected in Russia.

RESUMEN

Twenty one strains of rubella virus were isolated in the Western Siberia during 2004-2006 epidemic period. Genotyping of isolated strains was performed by partial sequencing of glycoprotein E1 gene. Phylogenetic analysis showed that 20 out of 21 isolated in the Western Siberia strains of rubella virus belonged to genotype 1g, and 1 strain (isolated in Altai region in 2006)--to genotype 1E.

RESUMEN

S-segment nucleotide sequences for two Crimean-Congo hemorrhagic fever (CCHF) virus strains isolated in the Rostov Region of Russia and in Bulgaria have been determined. Analysis of complete S-segment nucleotide sequences in the viral strains from different regions of the world has established that the CCHF virus strains isolated from ticks and human beings in different southern Russian regions in 1967 and 2000 are very closely genetically and they form an individual subgroup in the basic European genetic group. By the S-segment structure, the CCHF virus strain isolated in Bulgaria in 1978 belongs to the same genetic group as a representative of its second subgroup. Analysis of the S-segment 3'-noncoding region suggests that the CCHF virus circulating in Europe, Central Asia, and China may have originated from one global focus of infection, including several CCHF virus genovariants. During evolution, fragmental exchange apparently occurred in the S-segment 3'-noncoding region as a result of homological recombination.

RESUMEN

Pulmonary tissues of red-grey voles, Clethrionomys rufocanus from Shkotovo District and Maritime Territory were investigated. rt-PCR was used to detect the hantavirus nt-222 M-segment genome and nt-403 mitochondrial DNA fragment. A new genetic variant of the PUUV virus named as "Shkotovo", that is different from other PUUV strains by 18-23%, was shown to be circulating in red-grey voles. A phylogenetic analysis denoted 2 PUUV subgroups with their rodent-host branching into C. graleolus and C. rufocanus.

RESUMEN

The "VectoHantivirus-ampli" test system based on rtPCR was shown as fitting the detection of virus RNA in blood samples of patients with hemorrhagic fever with renal syndrome (HFRS) made at early stages and no later than 7 days from the disease onset. A sequence analysis of viral nucleotide sequences of PCR products produced by the test-system ensured the identification of Hantaviruses (pathogens of HFRS). Two genetic variants of Puumula virus were shown to circulate in the territory of the Bashkortostan Republic; they differ by 10.0-13.8%, one of them is absolutely new. The Hantan virus FE genetic variant was detected in the studied samples from the Khabarovsk Territory.

RESUMEN

Complete S-segment nucleotide sequences of genomic RNA were determined for two Crimea-Congo hemorrhagic fever (CCHF) virus strains, i.e. LEIV 10145 Uz isolated from ticks in Uzbekistan, 1985, and LEIV 29223 Stv isolated from a patient in Stavropol region, 2000. It was established that the S-segment length is 1672 and 1674 nucleotides. Therefore, the initiating codon (for methionine) is located at positions 56-58; the length of translation frames for the nucleocapsid protein is 482 amino acid residues. Distinctions in the length of S-segment, as compared to other strains, are related only with the 5' and 3' non-coding regions. A comparison of the nucleotide and amino-acid sequences of S-segments of genome of the mentioned strains with the early published data showed that the CCHF virus strain isolated in Uzbekistan is mostly close to strains isolated in China, and that the strain isolated in Stavropol region forms, jointly with Drozdov strain isolated in the Astrakhan region, a separate branch in the phylogenetic tree.

RESUMEN

Data on the circulation of a new genetic variant of Seol virus, which was designated as "Vladivostok" (the name of the place where it was isolated)--VDV, were obtained, by using the antigenic and genetic analysis of hantavirus strains isolated from patients with hemorrhagic fever with renal syndrome (HFRS) and from brown rats, and described in the present paper. A complete identity of nucleotide sequences of the M-segment fragment of isolates obtained from HFRS patients and of those obtained from rats was demonstrated, which confirms the key role of brown rat as a source of infection.

Five antigen-positive samples isolated from patients with Crimean-Congo hemorrhagic fever (CCHF) and from Hyalomma marginatum ticks collected in the European part of Russia and three laboratory strains of CCHF isolated in Russia, Uzbekistan, and Tadjikistan were analyzed by reverse transcription-polymerase chain reaction (RT-PCR) and sequencing. Comparison of nucleotide sequences of fragments of CCHF virus genome S segment and phylogenetic analysis of Russian strains showed that all CCHF strains isolated from humans and H. marginatum circulating in Russia were closely related and differed essentially from CCHF variants from other regions. Strains isolated in Uzbekistan and Tadjikistan were most closely related to CCHF strains from China.

Sera samples from patients suspected of Crimean-Congo hemorrhagic fever (CCHF) taken during epidemic outbreak at the territory of Sarysusky and Moiynkumsky districts of the Zhambyl region in Kazakhstan, in 2000, were analysed by means of reverse transcription-polymerase chain reaction (RT-PCR) and sequencing of virus genome fragments. Genome RNA of CCHF virus was found in 2 assays. Analysis of nucleotide sequences of fragments of S-segment of viral genome revealed in the Sarysusky districts circulation of CCHF virus, genetically resembled to close phylogenetically to CCHF virus strains from China.

RESUMEN

The nonstructural 36K protein of vaccinia virus (VV) mapped in HindIII-P and HindIII-J fragments of VV strain L-IVP has been expressed in E. coli as a fusion protein. The products of 36K gene preserved the antigen homology with the native protein 36K and the capacity to bind specific immunoglobulins of rabbit antiVV serum. The protective properties of 36K gene products and their joint effect with the immunodominant protein 35K were investigated. The non-structural 36K gene products showed no protective activity, but increased the production of specific antibodies in mice immunized with a mixture of both protein preparations, this increase being compatible with that observed after immunization with the inactivated virus preparations.

With regard to acute toxicity, the cubosols are attributed to the non-cumulative compounds of low toxicity. Acute inhalation exposure threshold for cubosol "Bright green" is over 140 mg/m3. Subacute poisonings developed following intragastric administrations in concentrations 1 g/kg for 30 days, and were characterized by polymorphism of the signs. Revealed were delayed body mass increment in the laboratory animals, changes in peripheral blood and hepatorenal morphology and function.

The article contains a morphofunctional evaluation of spermatogenic epithelium state in rats after chronic inhalation of Orange-25 disperse azo dye. The result of the study revealed gonadotoxic effects at both concentration levels 28.5 mg/m3 and 5.6 mg/m3, which manifested themselves through violations in the spermatozoa functional states and changes in the seminiferous epithelium morphometric indices. Judging from the affection of the blood, the concentration 28.5 mg/m3 can be considered toxic, and 5.6 mg/m3 can be regarded liminal. Thus, the gonadotoxic effect was a secondary one, as in male rats in both groups, a hemotoxic effect was revealed at toxic and liminal dosage concentrations.

Models of processive and distributive DNA synthesis and degradation catalysed by matrix enzymes were investigated. Distribution of polymer products dependent on the reaction model chosen, on the type of the matrix and on the enzyme-matrix initial concentration ratio were determined by methods of numerical modeling. Conditions were found where the scattering of the reaction polymer products was minimal. The homopolymer matrix choice of experimental condition may generate a distribution of product that obeys the Poisson distribution. Numerical investigations of polymerization (hydrolysis) processes showed that for a number of heteropolymer matrixes a distribution may exist with scattering much less than that for homopolymer matrixes. Conditions are found when processive and distributive models give different distributions of the reaction product.

Kinetic values of the BamHI endonuclease interaction with synthetic oligonucleotides, containing some defects, have been determined. These defects were: the absence of the one internucleotide phosphate in the GGATCC sequence; substitution of a phosphate linkage by a methylphosphonate one; 5'-protruding end of the double-stranded oligonucleotide substrate. Some modifications resulted in the increase of the initial rates of cleavage due to higher Vmax values for these substrates. Several structural defects in the oligonucleotide substrates have been shown to intensity the formation of productive complexes with the enzyme, which can be explained by the significant role of the polynucleotide chain kinks in the recognition process. Studies on oligonucleotides with different defects made it possible to reveal the phosphate groups essential for the interaction with BamHI endonuclease.

Interaction of Ecodam methylase (E.C. 2.1.1) with synthetic oligonucleotide substrates of various primary structure was studied by the small angle X-ray scattering method. Complex formation between the enzyme and substrates occurs after addition of double-stranded oligonucleotides to the methylase. In the presence of 1 M NaC1 (when the enzyme is inactive) addition of the synthetic substrates does not result in complex formation. Comparison of the experimental scattering parameters with the calculated ones has been made. The best coincidence of these data is obtained for the model which proposed Ecodam methylase dimer formation in the course of its interaction with the substrates.

The specificity of DNA X dye binding was studied. Antibiotic distamycin A was bound most strongly to the DNA sequences composed of three or more neighboring A X T pairs. Acrichin and 7-aminoacrichin proved to be weak specific inhibitors binding predominantly within the A X T regions.
