RESUMEN
Complete nucleotide sequence of genomic RNA of hepatitis A virus (HAV) rapidly replicating strain MB-7 was determined. Comparison of nucleotide and deduced amino-acid sequences demonstrated the highest level of identity of MB-7 with strain HAS-15 (above 99%) and high homology with other HAV strains (HM 175/7, CR326, and GBM/HFS) used in production of anti-hepatitis A vaccines. MB-7 was classified as subgenotype IA. Phylogenetic analysis showed that MB-7 is most closely related to the strain HAS-15 and the HAV variants circulating in Russia. Comparative analysis of genomic differences between MB-7 and HAS-15 with other HAV strains revealed among changes characteristic of MB-7 those typical of the described earlier rapidly replicating HAV strains (nt. 149-162 in 5'-untranslated region and changes in the VP3 and 2C genes). These results suggest the functional importance of changes in above-mentioned regions of HAV genome for the increased replication level of MB-7 in vitro.

The influence of a vaccine based on the MB-7 strain of hepatitis A virus (VP-MB-7) designed at the "Vector" Center of Virology and Biotechnology was studied. VP-MB-7 was found to provoke no allergic response and to have an activating effect on the specific and non-specific responses of cell and humoral immunity similar to those evoked by hepatitis A vaccine "Hep-A-in Vac".

A study was undertaken to prove that a vaccine drug based on the fast-growing MB-7 (MB-7 VD) of hepatitis A virus (HAV) is safe. It was established that MB-7 VD, when once injected to white outbred mice and when 4 times injected to Hartley guineas pigs, did not cause any hematological and biochemical changes in the peripheral blood or in the condition of the central nervous system of experimental animals, which shows that MB-7 VD is free of any toxic properties.

The fraction obtained from acidic extract of bovine brain homogenate after several steps of chromatographic purification provokes spontaneous aggressive encounters in rats upon intracerebroventricular injection. The simultaneous long-term raising of electric shock-induced aggression with the suppressing of muricidal and intraspecies aggressive behaviour has been observed. Intravenous and intraperitoneal injections of this fraction induce no behavioural changes in rats. It has been determined that the fraction consists of complex compounds of zinc with various aliphatic amines. A similar or higher behavioural activity has been discovered in series of synthetic complexes of zinc with different ligands, that are suggested for use in modelling any nervous and psychiatric disorders connected with an increased aggression level.

The presented software package allows: 1) to input and edit amino acid sequences; 2) to list aligned amino acid sequences of homologous proteins; 3) pairwise comparison of homologous sequences; 4) construction of phylogenetic trees; 5) comparison of two groups of protein sequences from the same family of homologous proteins; 6) graphic identification of conservative and variable regions of homologous sequences. The stepwise application of the programs allows to study the process of amino acid replacement accumulation during certain intervals of species evolution.

Earlier we found that the immune response and antiviral protection from FMDV can be achieved by immunization with uncoupled FMDV peptides. In a search of approaches to animal protection from FMDV A22 strain we prepared a series of peptides corresponding to the putative antigenic determinants. Synthetic 131-149 and 140-149 sequences afforded 50 to 80% protection, both in the free state and conjugated with keyhole limpet hemocyanin. We believe that the 140-149 segment is so far the smallest peptide capable of eliciting specific antiviral protection without conjugation with a high molecular carrier.

Fractionated soft gels Sephadex LH-20, Sephadex LH-60, Enzakryl K-2 and semirigid gels - Spheron P-40, Spheron P-300 and Spheron P-1000 were tested for separation of protected synthetic fragments of neurotoxin II from the Central Asian cobra (Naja naja oxiana) venom. Applicability of the above gels for size exclusion HPLC of protected peptides in organic media is discussed.
