RESUMEN

Cyanobacteria are able to synthesize a high diversity of natural compounds that account for their success in the colonization of a variety of ecological niches. Many of them have beneficial properties. The mud from the thermal baths of Balaruc-Les-Bains, one of the oldest thermal baths in France, has long been recognized as a healing treatment for arthro-rheumatic diseases. To characterize the cyanobacteria living in these muds, several strains were isolated from the water column and biofilms of the retention basin and analyzed using a polyphasic approach. Morphological, ultrastructural and molecular (16S rRNA gene and 16S-23S ITS region sequencing) methods were employed to identify nine cyanobacterial strains belonging to the orders Chroococcales, Synechococcales, Oscillatoriales and Nostocales. The combination of morphological and genetic characteristics supported the description of a new genus and species with the type species as Pseudochroococcus coutei. The taxonomic diversity in the muds from Thermes de Balaruc-Les-Bains appears higher than previously documented, providing new candidate taxa for their observed therapeutic properties.

High garlic dose could exert adverse health properties and grape seed and skin extract (GSSE) exhibit a variety of beneficial effects, even at high dose. In the present study we evaluated the toxic effect of high garlic dose treatment on antioxidant status of the blood compartment and the protective effect of GSSE. Rats were intraperitoneally (i.p.) administered either with garlic extract (5 g/kg bw) or GSSE (500 mg/kg bw) or a combination of garlic and GSSE at the same doses daily during one month. Plasma parameters and erythrocytes antioxidant status were evaluated. Data confirmed that high garlic dose induced anemia and a pro-oxidative state into erythrocytes characterized by increased malondialdehyde (MDA), carbonyl protein and antioxidant enzyme activities as catalase (CAT), peroxidase (POD) and superoxide dismutase (SOD). Garlic also elevated intracellular hydrogen peroxide (H(2)O(2)) and free iron whereas GSSE treatment counteracted almost all garlic deleterious effects. In conclusion, high garlic dose induced a pro-oxidative state into erythrocytes via the Fenton reaction between H(2)O(2) and free iron, and GSSE exerted antioxidant properties.

Doxorubicin (Dox), a widely used antitumor anthracycline antibiotic, plays an undisputed key role in the treatment of many neoplasic diseases. In this study, the protective role of resveratrol against Dox-induced erythrocytes and plasma toxicity has been evaluated in rats. Animals were treated with resveratrol (25 mg/kg b.w.) by intraperitoneal injection during 8 days. At the 4(th) day of treatment, rats were intraperitoneally injected with a single dose of Dox (20 mg/kg b.w.). At the end of the treatment, blood samples were collected following standard procedure and processed for oxidative stress parameters (malondialdehyde (MDA), carbonyl protein, free iron, calcium and H(2)O(2) levels), transaminases and antioxidant enzymes as catalase (CAT), peroxidase (POD) and superoxide dismutase (SOD). Data showed that Dox drastically increased erythrocytes and plasma MDA, free iron, H(2)O(2) and carbonyl proteins but decreased calcium levels and also decreased erythrocytes CAT, POD and SOD activity. Besides, Dox decreased plasma CAT and SOD but unexpectedly increased POD activity. Dox also increased plasma ALT and AST levels and decreased them into erythrocytes. Co-treatment with resveratrol counteracted almost all Dox's effects. In conclusion, Dox induced a pro-oxidative stress into erythrocytes and resveratrol exerted real antioxidant properties which can be attributed, at least in part, to free iron and calcium modulation.

Although microscope analysis is very useful for studying phytoplankton community composition, it does not allow for high frequency (spatial and/or temporal) data acquisition. In an attempt to overcome this issue, fluorescence-based approaches that use selective excitation of pigment antennae have spread rapidly. However, the ability of spectral fluorescence to provide accurate estimates of phytoplankton biomass and composition is still debated, and only a few datasets have been tested to date. In this study, we sampled of a wide range of water bodies (n=50) in the Ile-de-France region (North Central France). We used the resulting extensive dataset to assess the ability of the bbe-Moldaenke FluoroProbe II (FP) to estimate phytoplankton community composition in lakes and reservoirs. We demonstrated that FP data yields better estimates of total phytoplankton biovolume than do spectrophotometric chlorophyll a measures and that FP data can be further corrected using the average chlorophyll a to biovolume ratio among phytoplankton groups. Overall, group-specific relationships between FP and biovolume data were consistent. However, we identified a number of cases where caution is required. We found that Euglenophytes are expected to depart from the global FP vs. biovolume relationship of the 'green' group due to varying Fv/Fm and pigment content in response to environmental conditions (photoautotrophic vs. photoheterotrophic growth). Then, it appears necessary to consider the composition of the Chromophytes community in order to obtain a good agreement between both biomass estimation methods. Finally, we confirmed the misattribution toward the 'red' group of phycoerythrin-containing cyanobacteria and the occurrence of a strong scattering in the relationship between the FP vs. biovolume of the 'blue' group that can be partly attributed to the occurrence of large colony-forming cyanobacteria (e.g., Microcystis spp, Aphanizomenon flos-aquae). We propose correcting procedures to improve the quality of data obtained from spectral fluorescence tools in the context of large-scale sampling of lakes and reservoirs.
