Decreased clearance of i.v. or intratracheally administered gentamicin was observed in rats following single whole-body irradiation. 6 Gy 60Co. reaching its lowest rate on the 7th post-irradiation day. Simultaneously the absorption rate of gentamicin from the lungs was found to be increased.

After i.v. administration to rats of polysaccharide-protein complex, isolated from Candida albicans, a decrease of cardiac output was observed from 20 sec to 240 min postinjection, followed by a recovery at 360 min. Concomitantly the regional blood flow was maintained in heart and lungs, moderately decreased in intestine, liver and adrenals and markedly reduced in skin, muscle, spleen and kidney.

The pharmacokinetics of intratracheally instilled and intravenously injected gentamicin were compared in the rat and analyzed by a one-compartment open model. The maximum concentration of gentamicin in plasma occurred within 10 min after intratracheal instillation. Considerable amounts of gentamicin were absorbed from lungs after intratracheal instillation, as shown by its concentrations in plasma and elimination in urine. The data suggest that the absorption of gentamicin from the pulmonary system would be sufficient to maintain therapeutic levels of this agent in plasma.

The organ distribution of 131I-o-iodohippurate (OIH) was studied in rats in the time interval of 1--1440 min. The mean decrease of OIH in plasma was fitted with a function composed of three exponential terms. A three-compartment model describing kinetics of OIH in rats was suggested and its parameters were computed. Comparison of organ and plasma concentrations of OIH showed that the liver, up to 30 min, and the heart and lungs, up to 15 min after administration, form with plasma a common central compartment.
