Background: Phyllodes tumors (PTs) account for 0.3-1.0% of all breast tumors and often occur in women aged 35 to 55. They are similar to giant fibroadenomas. PTs are famous for local recurrence. No more than 10% of PTs grow larger than 10 cm. The National Comprehensive Cancer Network (NCCN) guidelines recommend extensive resection with a margin of ≥1 cm for PTs, which is much larger than that required for breast cancer. Positive resection margin is associated with recurrence. However, little is known about whether all subtypes really require radical tumor negative resection margins. Case Description: We report on a 49-year-old woman with a giant borderline PT in her left breast. The tumor was greater than 10.5 cm × 7.0 cm. She had a bilateral benign PT excision in January 2014 and a left benign PT excision in December 2018. A chest computerized tomography (CT) scan and abdomen ultrasound did not reveal distant metastasis. Therefore, left breast mastectomy was performed. Wound healing was satisfactory. Pathological and immunohistochemistry findings showed a borderline PT. Conclusions: As the rare tumor of the breast, PTs pose a great challenge for surgeons. The initial evaluation of PTs relies on a triple evaluation of clinical, radiological, and histological examination. Local recurrence of PTs is more common than distant metastasis. The histology of recurrent tumors is usually identical to that of the primary tumor, or has a tendency to malignancy. Although most surgeons are uncomfortable with PTs with a positive margin, it is reasonable to adopt a "watchful waiting" strategy for benign PTs. The current recommendation that PTs should be extensively resected regardless of tumor size might be revised.

In this work, the two-dimensional self-assembly and co-assembly behaviors of two tetracarboxylic acid derivatives (H4BDETP and H4BTB) were investigated by scanning tunneling microscopy (STM). H4BDETP molecules self-assembled into linear nanostructures, and H4BTB molecules formed lamellar and tetragonal nanostructures. The formation of a H4BDETP/H4BTB co-assembly nanostructure was closely related to the deposition sequence of H4BDETP and H4BTB on highly oriented pyrolytic graphite (HOPG). The introduction of H4BTB into the self-assembly system of H4BDETP resulted in the emergence of the H4BDETP/H4BTB nanostructure, while the addition of H4BDETP had no effect on the self-assembly system of H4BTB and a H4BDETP/H4BTB co-assembly nanostructure was not obtained.

The supramolecular self-assembly behavior of a pair of low-symmetry tetracarboxylic acid molecules (H4OBDB and H4ADDI) and their co-assembly behavior with TMA as a bridging molecule were studied at the liquid-solid interface. Scanning tunneling microscope (STM) observations revealed that H4OBDB and H4ADDI molecules both tend to form O-shaped dimers but end up forming different types of self-assembly structures. We also investigated the construction of two-component co-assembly structures by mixing H4OBDB or H4ADDI molecules with bridging molecules such as TMA. The two formed co-assembly structures are similar. Based on the analysis of the STM results and the density functional theory (DFT) calculations, the formation mechanism of the assembled structures was revealed.

Based on previous research, a new coassembly formed by a porphyrin derivative (IPETPP), which contains a flexible substituent of m-phthalic acid, is observed with coronene (COR) molecules at a higher concentration. Besides, a fresh IPETPP self-assembly formed at a lower concentration and another new coassembly with COR molecules are obtained. Moreover, the addition of a series of bipyridines alters the diamond arrangement of IPETPP, which enhances the stability of the two-component structures. It is unprecedented that bipyridine derivatives break intermolecular hydrogen bonds containing m-phthalic acid substituents. All the coassemblies are investigated by scanning tunneling microscopy on a highly oriented pyrolytic graphite. Combined with density functional theory, the formation mechanism of the assembled structures is revealed. These results would contribute to understanding the interfacial crystal behaviors and probably provide an efficient pathway to regulate the binary structures.

BACKGROUND: This study aimed to investigate the clinicopathological significance of sine oculis homeobox homolog 1 (SIX1) and eyes absent 1 (EYA1) in patients with chronic hepatitis B (CHB) and other liver diseases. METHODS: SIX1 and EYA1 levels were detected in human serum and liver tissues by enzyme linked immunosorbent assay (ELISA) and immunofluorescent staining method, respectively. RESULTS: The serum SIX1 and EYA1 levels in 313 CHB patients were 7.24±0.11 and 25.21±0.51 ng/mL, respectively, and these values were significantly higher than those in 33 healthy controls (2.84±0.15 and 13.11±1.01 ng/mL, respectively; P<0.05). Serum SIX1 and EYA1 levels were also markedly increased in patients with numerous other liver diseases, including liver fibrosis, hepatocellular carcinoma, fatty liver disease, alcoholic liver disease, fulminant hepatic failure, autoimmune liver disease, and hepatitis C, compared to the healthy controls (P<0.05). Dynamic observation of these proteins over time in 35 selected CHB patients revealed that SIX1 and EYA1 serum levels increased over an interval. Immunofluorescent staining revealed that both SIX1 and EYA1 were only expressed in hepatic stellate cells (HSCs), and their increased expression was evident in CHB liver tissue. CONCLUSIONS: SIX1 and EYA1 are novel biomarkers of liver damage in patients of CHB and other liver diseases, with potential clinical utility.

OBJECTIVE: α-1 antitrypsin deficiency (AATD) is an inherited liver disease characterized by the "Z" mutations, which can cause pulmonary emphysema and liver fibrosis. Transplantation of the organ (i.e., the lung/liver) is the best treatment method, however, the scarcity of suitable donors limits its application. The cell transplantation technique poses an alternative way of combating liver failure. METHODS: Hepatic specific differentiation of the human induced pluripotent stem cells (iPSCs) was initiated with 100 ng/mL activin A, followed by 20 ng/mL of BMP-4 and 10 ng/mL of FGF-2. The cells were transplanted into the livers of AATD transgenic mice using intra-splenic injections. FK506 was used as an immunosuppressor. At 1, 3, and 6 months post-transplantation, the human serum albumin (HSA) levels and its DNA contents, and the mice serum and liver tissues were measured using enzyme-linked immunosorbent assays (ELISA), polymerase chain reactions (PCR), and immunohistochemistry to estimate the repopulation of the hepatic-specified cells. RESULTS: Post transplantation, the hepatic-specified cells were found to be successfully and progressively repopulated in the transgenic mice livers. Additionally, the hepatic-specified cells did not display any carcinogenicity, as confirmed by the absence of any tumors on the animals. CONCLUSION: We provide a time saving and low cost method of transplanting hepatic-specified cells into the livers of AATD mice without any risk of carcinogenicity, a method that may be a potential option for the treatment of AATD.

α-Ketoglutarate (α-KG), a pivotal metabolite in energy metabolism, has been implicated in nonalcoholic fatty liver disease (NAFLD) and several cancers. It is recently proposed that plasma α-KG is a surrogate biomarker of NAFLD. Here, we report the development of a novel "turn-on" chemosensor for α-KG that contains a coumarin moiety as a fluorophore. Using benzothiazole-coumarin (BTC) as inspiration, we designed a probe for calcium ion recognition that possesses a unique fluorophore compared with previously reported probes for α-KG measurement. This chemosensor is based on the specific Schiff base reaction and the calcium ion recognition property of the widely used calcium indicator BTC. The probe was synthesized, and a series of parallel experiments were conducted to optimize the chemical recognition process. Compared to the initial weak fluorescence, a remarkable 7.6-fold enhancement in fluorescence intensity (I/I0 at 495 nm) was observed for the conditions in which the probe (1 µmol/L), α-KG (50 µmol/L), and Ca2+ (100 µmol/L) were incubated at 30 °C in EtOH. The probe displayed good selectivity for α-KG even in an environment with an abundance of amino acids and other interfering species such as glutaric acid. We determined that the quantitative detection range of α-KG in EtOH was between 5 and 50 µmol/L. Finally, probe in serum loaded with α-KG (10 mmol/L) showed a 7.4-fold fluorescence enhancement. In summary, a novel probe for detecting the biomarker α-KG through a typical Schiff base reaction has been discovered. With further optimization, this probe may be a good alternative for detecting the physiological metabolite α-KG.

A simple, rapid and reliable spectrophotometry was developed to determine monoamine oxidase (MAO). In this study, 2,4-dinitrophenylhydrazine (DNPH), a classic derivatizing reagent, was used to detect MAO-dependent aldehyde production; and traditional DNPH spectrophotometry was simplified. Benzylamine and serotonin oxidation were catalyzed by MAO-B and MAO-A, respectively, to aldehydes. These were derivatized with DNPH, and the corresponding quinones were further formed by adding NaOH. These DNPH derivatives with large conjugated structures were directly measured spectrophotometrically at 465 nm and 425 nm, without the need for precipitating, washing and suspending procedures. The addition of NaOH caused a red shift of the maximum absorption wavelength of these derivatives, which reduced the interference of free DNPH. MAO-B protein was as low as 47.5 µg in rat liver with correlation coefficients ranging within 0.995-0.999. This method is 2-3 times more sensitive than direct spectrophotometry. The detection of MAO inhibition through this method showed that IC50 values of rasagiline are 8.00 × 10(-9) M for MAO-B and 2.59 × 10(-7) M for MAO-A. These results are similar to the values obtained by direct spectrophotometry. Our study suggests that DNPH spectrophotometry is suitable to detect MAO activity, and has the potential for MAO inhibitor screening in the treatment of MAO-mediated diseases.

Combined effects of enrichment procedure and non-fermentable acetate or fermentable glucose on system performance and bacterial community for pentachlorophenol (PCP) degradation in microbial fuel cells (MFCs) were determined in this study. Co-substrate and PCP were added into MFCs either simultaneously or sequentially. Simultaneous addition with glucose (simultaneous-glucose) achieved the shortest acclimation time and the most endurance to heavy PCP shock loads. Species of Alphaproteobacteria (simultaneous-acetate, 33.9%; sequential-acetate, 31.3%), Gammaproteobacteria (simultaneous-glucose, 44.1%) and Firmicutes (sequential-glucose, 31.8%) dominated the complex systems. The genus Sedimentibacter was found to exist in all the cases whereas Spirochaetes were merely developed in simultaneous-acetate and simultaneous-glucose. While Epsilonproteobacteria were only absent from sequential-acetate, simultaneous-glucose benefited to the evolution of Lentisphaerae. These results demonstrate simultaneous-glucose is a strategy for efficient system performance and the microbiological evidence can contribute to improving understanding of and optimizing PCP degradation in MFCs.

The combined anaerobic-aerobic conditions in air-cathode single-chamber MFCs were used to completely mineralize pentachlorophenol (PCP; 5 mg/L), in the presence of acetate or glucose. Degradation rates of 0.140 ± 0.011 mg/L-h (acetate) and 0.117 ± 0.009 mg/L-h (glucose) were obtained with maximum power densities of 7.7 ± 1.1 W/m(3) (264 ± 39 W/m(2), acetate) and 5.1 ± 0.1 W/m(3) (175 ± 5 W/m(2), glucose). At a higher PCP concentration of 15 mg/L, PCP degradation rates increased to 0.171 ± 0.01 mg/L-h (acetate) and 0.159 ± 0.011 mg/L-h (glucose). However, power was inversely proportional to initial PCP concentration, with decreases of 0.255 W/mg PCP (acetate) and 0.184 W/mg PCP (glucose). High pH (9.0, acetate; 8.0, glucose) was beneficial to exoelectrogenic activities and power generation, whereas an acidic pH = 5.0 decreased power but increased PCP degradation rates (0.195 ± 0.002 mg/L-h, acetate; 0.173 ± 0.005 mg/L-h, glucose). Increasing temperature from 22 to 35°C enhanced power production by 37% (glucose) to 70% (acetate), and PCP degradation rates (0.188 ± 0.01 mg/L-h, acetate; 0.172 ± 0.009 mg/L-h, glucose). Dominant exoelectrogens of Pseudomonas (acetate) and Klebsiella (glucose) were identified in the biofilms. These results demonstrate that PCP degradation using air-cathode single-chamber MFCs may be a promising process for remediation of water contaminated with PCP as well as for power generation.

Pentachlorophenol (PCP) was more rapidly degraded in acetate and glucose-fed microbial fuel cells (MFCs) than in open circuit controls, with removal rates of 0.12 ± 0.01 mg/Lh (14.8 ± 1.0 mg/g-VSS-h) in acetate-fed, and 0.08 ± 0.01 mg/L h (6.9 ± 0.8 mg/g-VSS-h) in glucose-fed MFCs, at an initial PCP concentration of 15 mg/L. A PCP of 15 mg/L had no effect on power generation from acetate but power production was decreased with glucose. Coulombic balances indicate the predominant product was electricity (16.1 ± 0.3%) in PCP-acetate MFCs, and lactate (19.8 ± 3.3%) in PCP-glucose MFCs. Current generation accelerated the removal of PCP and co-substrates, as well as the degradation products in both PCP-acetate and PCP-glucose reactors. While 2,3,4,5-tetrachlorophenol was present in both reactors, tetrachlorohydroquinone was only found in PCP-acetate MFCs. These results demonstrate PCP degradation and power production were affected by current generation and the type of electron donor provided.
