RESUMEN

Staphylococcus aureus is generally thought to divide in three alternating orthogonal planes over three consecutive division cycles. Although this mode of division was proposed over four decades ago, the molecular mechanism that ensures this geometry of division has remained elusive. Here we show, for three different strains, that S. aureus cells do not regularly divide in three alternating perpendicular planes as previously thought. Imaging of the divisome shows that a plane of division is always perpendicular to the previous one, avoiding bisection of the nucleoid, which segregates along an axis parallel to the closing septum. However, one out of the multiple planes perpendicular to the septum which divide the cell in two identical halves can be used in daughter cells, irrespective of its orientation in relation to the penultimate division plane. Therefore, division in three orthogonal planes is not the rule in S. aureus.

Carbohydrates from plant cell walls are often found as heteropolysaccharides intertwined with each other. For competitive advantage against other microorganisms, and ability to fully exploit available carbon and energy sources, Bacillus subtilis possesses a high number of proteins dedicated to the uptake of mono- and oligosaccharides. Here, we characterize transporter complexes, belonging to the ATP-binding cassette (ABC) superfamily, involved in the uptake of oligosaccharides commonly found in pectin. The uptake of these carbohydrates is shown to be MsmX-dependent, assigning a key role in pectin mobilization for MsmX, a multipurpose ATPase serving several distinct ABC-type I sugar importers. Mutagenesis analysis of the transmembrane domains of the AraNPQ MsmX-dependent importer revealed putative residues for MsmX interaction. Interestingly however, although MsmX is shown to be essential for energizing various ABC transporters we found that a second B. subtilis ATPase, YurJ, is able to complement its function when placed in trans at a different locus of the chromosome.

Area monitoring is a fundamental test at radiodiagnostic facilities to maintain an acceptable level of radiation exposure for employees and members of the public. Experimental measurements were taken in an ionising radiation calibration laboratory. Four area monitor instruments were used. Dose and dose rate measurements were measured in integrated and rate operating modes. The results show that precautions are necessary where the area monitor uses exposure times of ≤2 s. When taking measurements in rate mode for times ≤2 s, the area monitors evaluated show a tendency to underestimate dose rate, the inaccuracy was 41 %, and varied between 34 and 45 % for different energies. It is highly recommendable to work in integrated mode, inaccuracy varied from 2 to 35 %. For measurements taken with exposure times of ≥3 s, the average inaccuracy was 15 % and the range was between 2 and 41 %.
