RESUMEN

Endophytic fungi have emerged as a significant source of natural products with remarkable bioactivities. Recent research has identified numerous antioxidant molecules among the secondary metabolites of endophytic fungi. These organisms, whether unicellular or micro-multicellular, offer the potential for genetic manipulation to enhance the production of these valuable antioxidant compounds, which hold promise for promoting health, vitality, and various biotechnological applications. In this study, we provide a critical review of methods for extracting, purifying, characterizing, and estimating the total antioxidant capacity (TAC) of endophytic fungi metabolites. While many endophytes produce metabolites similar to those found in plants with established symbiotic associations, we also highlight the existence of novel metabolites with potential scientific interest. Additionally, we discuss how advancements in nanotechnology have opened new avenues for exploring nanoformulations of endophytic metabolites in future studies, offering opportunities for diverse biological and industrial applications.

RESUMEN

Due to their size, flexibility, biocompatibility, large surface area, and variable functionality nanoparticles have enormous industrial, agricultural, pharmaceutical and biotechnological applications. This has led to their widespread use in various fields. The advancement of knowledge in this field of research has altered our way of life from medicine to agriculture. One of the rungs of this revolution, which has somewhat reduced the harmful consequences, is nanotechnology. A helpful ingredient for plants, silicon (Si), is well-known for its preventive properties under adverse environmental conditions. Several studies have shown how biogenic silica helps plants recover from biotic and abiotic stressors. The majority of research have demonstrated the benefits of silicon-based nanoparticles (Si-NPs) for plant growth and development, particularly under stressful environments. In order to minimize the release of brine, heavy metals, and radioactive chemicals into water, remove metals, non-metals, and radioactive components, and purify water, silica has also been used in environmental remediation. Potentially toxic elements (PTEs) have become a huge threat to food security through their negative impact on agroecosystem. Si-NPs have the potentials to remove PTEs from agroecosystem and promote food security via the promotion of plant growth and development. In this review, we have outlined the various sources and ecotoxicological consequences of PTEs in agroecosystems. The potentials of Si-NPs in mitigating PTEs were extensively discussed and other applications of Si-NPs in agriculture to foster food security were also highlighted.

Advances in molecular pharmacology and an improved understanding of the mechanism of most diseases have created the need to specifically target the cells involved in the initiation and progression of diseases. This is especially true for most life-threatening diseases requiring therapeutic agents which have numerous side effects, thus requiring accurate tissue targeting to minimize systemic exposure. Recent drug delivery systems (DDS) are formulated using advanced technology to accelerate systemic drug delivery to the specific target site, maximizing therapeutic efficacy and minimizing off-target accumulation in the body. As a result, they play an important role in disease management and treatment. Recent DDS offer greater advantages when compared to conventional drug delivery systems due to their enhanced performance, automation, precision, and efficacy. They are made of nanomaterials or miniaturized devices with multifunctional components that are biocompatible, biodegradable, and have high viscoelasticity with an extended circulating half-life. This review, therefore, provides a comprehensive insight into the history and technological advancement of drug delivery systems. It updates the most recent drug delivery systems, their therapeutic applications, challenges associated with their use, and future directions for improved performance and use.

The etiopathogenesis of inflammatory and autoimmune diseases, including pulmonary disease, atherosclerosis, and rheumatoid arthritis, has been linked to human exposure to volatile organic compounds (VOC) present in the environment. Chronic inflammation due to immune breakdown and malfunctioning of the immune system has been projected to play a major role in the initiation and progression of autoimmune disorders. Macrophages, major phagocytes involved in the regulation of chronic inflammation, are a major target of VOC. Excessive and prolonged activation of immune cells (T and B lymphocytes) and overexpression of the master pro-inflammatory constituents [cytokine and tumor necrosis factor-alpha, together with other mediators (interleukin-6, interleukin-1, and interferon-gamma)] have been shown to play a central role in the pathogenesis of autoimmune inflammatory responses. The function and efficiency of the immune system resulting in immunostimulation and immunosuppression are a result of exogenous and endogenous factors. An autoimmune disorder is a by-product of the overproduction of these inflammatory mediators. Additionally, an excess of these toxicants helps in promoting autoimmunity through alterations in DNA methylation in CD4 T cells. The purpose of this review is to shed light on the possible role of VOC exposure in the onset and progression of autoimmune diseases.

The vast application of hemicellulose in industry is greatly influenced by its chemical components. The current study focuses on identifying the chemical components of a high yield alkaline-extracted hemicellulose and characterization to serve as a guide for more specific and effective biotechnological uses. In this study we isolated hemicellulose from sawdust of three different wood species (Beech, African rose and Agba woods) and characterized them using FTIR and HPLC techniques. Hydroxyl spectra vibrations were observed at 3919-3671 cm-1 and 3454-3211 cm-1 which indicates the presence of non-hydrogen bonded OH stretch and normal polymeric OH stretch in all three samples. The samples contained residual lignin indicated by IR absorption bands at 1592 and 1525 cm-1. The presence of C=O stretching vibrations of acetyl groups at 1734 cm-1 indicated that African rosewood was generally an acetylated molecule. Each heteropolysaccharide also contained reducing monosaccharides at their ends suggested by the C-H stretching vibrations. Infrared absorptions characteristic of asymmetric ß-1,6-glycosidic stretching was present in Beechwood and Agbawood, respectively, and African rosewood gave three absorption bands ß-1,3-glycosidic stretch, ß-1,4-glycosidic stretch and an asymmetric ß 1,6-glycosidic stretch, respectively. Agbawood gave a major absorption band at 923.75 cm-1 corresponding to the absorption band at ß-1,4-glycosidic stretching. African rosewood contained 96 % mannose and 4 % of an unidentified sugar. Beechwood contained primarily glucose, but Agbawood contained 20, 14, 8 and 57 % glucose, mannose, galactose, and an unidentified sugar, respectively.

The molecular screening for laccase specific gene sequences in Trametes polyzona WRF03 (TpWRF03) using designed oligonucleotide primers analogous to the conserved sequences on the copper-binding regions of known laccases showed positive amplification with an amplicon size corresponding to 1500 bp. The purified TpWRF03 laccase (TpL) is a monomer with a molecular weight corresponding to 66 kDa. The enzyme had an optimal pH of 4.5 and temperature of 55 °C. TpL was most stable within pH of 5.5-6.5 and at a temperature range of 40-50 °C. Sodium azide, sodium cyanide and Fe2+ greatly inhibited the enzyme activity. TpL showed more than 50 % decolourisation efficiency on coomassie brilliant blue (72.35 %) and malachite green (57.84 %) but displayed low decolourisation efficiency towards Azure B (1.78 %) and methylene blue (0.38 %). The results showed that TpWRF03 produces high-yield of true laccase with robust properties for biotechnological applications.
