RESUMEN
The pregnant rats were treated with formaldehyde (0.5 mg/kg daily per os) during whole period of pregnancy. The activity of cytochrome-c-oxidase, malate dehydrogenase, nucleotidase, glucose-6-phosphatase, beta-glucuronidase, N-acetyl-beta-glucosaminidase, beta-galactosidase, H(+)-ATPase, glutamate dehydrogenase, NAD- and NADP-isocitrate dehydrogenase, fructose-bisphosphate aldolase, glucose-6-phosphate dehydrogenase and content of protein in liver celts of offsprings (newborns, 2 weeks age and 2 months age) were studied. It was shown differences in development enzyme systems of control and experimental animals during ontogenesis.

Functional state of the system involved in biotransformation of xenobiotics in animal liver tissue was studied under conditions of combined chemical contamination of atmospheric air and water simulating the typical real loading. The model loading studied caused in some cases an induction of the microsomal monooxygenases without activation of UDP-glucuronosyl transferase, but simultaneously with accumulation of malonic dialdehyde; this suggests that metabolic alterations observed may be involved in development of pathology.

Dissimilar response of rat kidney microsomal and mitochondrial enzymes was observed after administration of nitrosodimethylamine at concentrations 0.1, 1.0 and 10.0 mg/L within 2, 5 and 10 months. Activity of the mitochondrial enzymes studied depended to a greater degree on concentration of the drug administered as compared with duration of its effect. At the same time, activity of the microsomal enzymes studied was increased depending on duration of nitroso-++ dimethylamine action administered at concentration of 0.1 mg/L, while it did not alter if concentration of the drug was increased from 1.0 to 10.0 mg/L within the all periods of examination.

Functional state of enzymatic systems in rat liver endoplasmic reticulum was studied. Content of cytochromes P-450 and b5 as well as activities of UDP-glucuronyl transferase, glucose-6-phosphatase, beta-glucuronidase, acetylesterase, inosine-5-diphosphatase were evaluated after permanent administration of nitrosodimethylamine within 2, 5 and 10 months at concentrations of 0.1, 1.0 and 10.0 mg/l. Content of the cancerogene active metabolites in liver cells was shown to be responsible for impairment of the functional state of microsomal enzymatic systems and was also related to intensity and duration of the nitrosodimethylamine effect. The level of these cancerogene metabolites in liver cells depended on rates of the cancerogene oxidation in the monooxygenase system and elimination of active products. At the same time, the rate of nitrosodimethylamine metabolism correlated with the doses of the substance administered into rats.

The results of a comparative biochemical and electron-microscopic study of membrane-injuring effect of N-nitrosodimethylamine--a chemical carcinogen widespread in the environment are reviewed. It has been established that in vivo and in vitro membrane-injuring effect of nitrosodimethylamine is characterized by reduced activity of membrane-bound enzymes of endoplasmic reticulum (UDP-glucuronyltransferase and glucose-6-phosphatase) accompanied by alterations in functional morphology of cells and intracellular structure.

Content of progesterone and cortisol was studied in blood plasma. Activities of organelle-specific enzymes were estimated in mitochondria (malate dehydrogenase, glutamate dehydrogenase, H + ATPase) and in endoplasmic reticulum (inosine-5-diphosphatase) of liver, kidney tissues and blood serum of rats, which were born after the influence of embryotoxic effectors. Correlation was found in alterations of the patterns studied. The steroid hormonal systems appear to affect the stability of mitochondrial and microsomal membranes in liver and kidney tissues of experimental animals.

Simultaneous effect of benz(a)pyrene and sulphurous gas was accompanied by distinct alteration in metabolism of sialoglycoproteins in liver tissue; at the time N-acetylneuraminic acid was redistributed in liver subcellular organelles (mitochondria, lysosomes and endoplasmic reticulum) as well as cytochromes P-450 and b5 were induced in liver microsomes. The alterations observed appear to play a definite role in manifestation of the unfavourable effect in combined action of the carcinogen and chemical contaminations of environments.
