RESUMEN

The occurrence of enhanced concentration of the radium triplet 226Ra, 228Ra and 224Ra is a frequently observed property of highly saline anoxic deep water as used e.g. in geothermal plants. In the present study we develop a model to explain the observed activity levels in the brines. The model considers processes at the rock-fluid interface of the aquifer like alpha recoil, sorption and surface precipitation and is implemented by means of a Monte Carlo simulation. The outcomes of the simulations indicate the dominating role of fine-grained constituents of the reservoir rock, e.g. claystone with enhanced specific activities of the natural decay chains. Mass fractions of such material in the order of a few percent are sufficient to result in radium fluid concentrations >1 Bq l-1. Also a generally valid relation between the Th/U ratio in the aquifer rock and the 228Ra/226Ra activity ratio in the fluid was found. This link improves the agreement between radium fluid data and the mean Th/U ratio of the Earth's crust. The 224Ra/228Ra fluid ratios reflect the transport time from the location of last radium release to the sampling point. The model findings were applied to a well investigated aquifer used in a geothermal plant in the North German Basin. An eight component system of the aquifer rock was established as the basis for the simulation of the radium concentrations in the deep fluid. The comparison between simulation and fluid analyses revealed a degree of radium sorption of about 50 %, which is necessary to match the model's results with the measurements. On the other hand, the 228Ra/226Ra fluid ratio of the brine was well reproduced by the simulation, showing the suitability of the model even in complex heterogeneous reservoirs. From the 224Ra/228Ra fluid ratios a transition from pore-to fracture-guided transport < 10 m distance from the production well is suggested. Precipitates from such deep fluids occurring after changes of the thermodynamic conditions are able to accumulate radium isotopes in Ba/Sr-sulphate phases. The time dependence of the radioactive disequilibrium between 226Ra, 228Ra and its child 228Th in such scales is described by a mathematical model and is applied to two different uptake models. Based on this approach, age determinations on precipitates found in different components of a geothermal plant are conducted. They reveal the triggering of scale formation due to modifications in the plant. The results are suitable for drawing conclusions about the operation of the system, which result in a reduction in the amount of scale and a reduction in downtimes.

The adhesion G-protein-coupled receptor CD97/ADGRE5 is present in adherens junctions of human normal intestinal cells and upregulated in colorectal carcinomas. Here, we examined whether CD97 directly interacts with junctional proteins in normal and malignant colorectal tissue. We identified an association of CD97 with ß-catenin using a proximity ligation assay and confirmed the interaction between both endogenous proteins at the biochemical level by co-immunoprecipitation in human and mouse tissues and cell lines. Glutathione S-transferase-pulldown revealed that CD97 binds ß-catenin through its seven-span transmembrane/intracellular domain(s). To study tumor-associated changes in the interaction of CD97 and ß-catenin in situ, we quantified and correlated both proteins at the membrane, and in the cytoplasm and nuclei of colorectal carcinomas and their corresponding normal tissues (n = 111). In normal colon, membranous levels of CD97 and ß-catenin correlated strongly (p < 0.0001). To some degree both molecules disappeared in carcinomas simultaneously from the membrane of tumor cells (p = 0.017). CD97 accumulated in the cytoplasm, whereas ß-catenin emerged in the cytoplasm and nuclei. CD97 and ß-catenin levels in the cytoplasm correlated well (p < 0.0001). Irrespective of their subcellular localization, interaction of CD97 with ß-catenin in tumor cells was also restricted to the cell contacts. Accordingly, CD97 did not regulate ß-catenin-dependent TCF-mediated transcriptional activity. In summary, while CD97 and ß-catenin interact in adherens junctions, their interaction is lost and both molecules follow different functional paths inside tumor cells.

At a progressive rate, small wastewater treatment plants in rural areas need to be equipped with an additional phosphorus removal stage in order to achieve a good chemical status in the receiving natural water bodies. A conventional regeneration method for ferric (hydr)oxides such as phosphate specific adsorbents, which can be applied to remove and recover phosphorus in fixed bed filters, was investigated and improved. It was shown that a loss of up to 85% of the initial capacity can be observed when regeneration with 1 M NaOH is implemented. The losses are caused by surface blocking with different calcium-containing compounds as revealed by an EDX analysis. These blocking compounds could be removed completely with an additional acidic regeneration step at pH = 2.5. During the alkaline desorption that followed, complete phosphorus removal and a full recovery of the adsorption capacity were achieved for goethite-rich Bayoxide(®) E 33 HC (E33HC) and akaganéite-rich GEH(®) 104 (GEH). The regeneration procedure was repeated up to eight times without any signs of further decline in the phosphate adsorption capacity or any changes in the specific surface area or pore size distribution of the adsorbent. In contrast to GEH and E33HC, ferric hydroxide- and calcite-rich FerroSorp(®) Plus (FSP) was partly dissolved during acid treatment.

Postnatal enlargement of the mammalian intestine comprises cylindrical and luminal growth, associated with crypt fission and crypt/villus hyperplasia, respectively, which subsequently predominate before and after weaning. The bipartite adhesion G protein-coupled receptor CD97 shows an expression gradient along the crypt-villus axis in the normal human intestine. We here report that transgenic mice overexpressing CD97 in intestinal epithelial cells develop an upper megaintestine. Intestinal enlargement involves an increase in length and diameter but does not affect microscopic morphology, as typical for cylindrical growth. The megaintestine is acquired after birth and before weaning, independent of the genotype of the mother, excluding altered availability of milk constituents as driving factor. CD97 overexpression does not regulate intestinal growth factors, stem cell markers, and Wnt signaling, which contribute to epithelial differentiation and renewal, nor does it affect suckling-to-weaning transition. Consistent with augmented cylindrical growth, suckling but not adult transgenic mice show enlarged crypts and thus more crypt fissions caused by a transient increase of the crypt transit-amplifying zone. Intestinal enlargement by CD97 requires its seven-span transmembrane/cytoplasmic C-terminal fragment but not the N-terminal fragment binding partner CD55. In summary, ectopic expression of CD97 in intestinal epithelial cells provides a unique model for intestinal cylindrical growth occurring in breast-fed infants.

The occurrence of the taste and odour compounds geosmin and 2-methyl isoborneol (2-MIB) affects the organoleptic quality of raw waters from drinking water reservoirs worldwide. UV-based oxidation processes for the removal of these substances are an alternative to adsorption and biological processes, since they additionally provide disinfection of the raw water. We could show that the concentration of geosmin and 2-MIB could be reduced by VUV irradiation and the combination of UV irradiation with ozone and hydrogen peroxide in pure water and water from a drinking water reservoir. The figure of merit EE/O is an appropriate tool to compare the AOPs and showed that VUV and UV/O(3) yielded the lowest treatment costs for the odour compounds in pure and raw water, respectively. Additionally, VUV irradiation with addition of ozone, generated by the VUV lamp, was evaluated. The generation of ozone and the irradiation were performed in a single reactor system using the same low-pressure mercury lamp, thereby reducing the energy consumption of the treatment process. The formation of the undesired by-products nitrite and bromate was investigated. The combination of VUV irradiation with ozone produced by a VUV lamp avoided the formation of relevant concentrations of the by-products. The internal generation of ozone is capable to produce ozone concentrations sufficient to reduce EE/O below 1 kWh m(-3) and without the risk of the formation of nitrite or bromate above the maximum contaminant level.
