RESUMEN

Cancer-associated fibroblasts (CAF) are a heterogeneous group of tumor microenvironment cells that are barely studied in metastatic lymph nodes. The presence of CAF in regional metastases of colorectal cancer was assessed by using SMA, PDGFRb, and POD markers; the obtained subpopulations were compared with the primary tumor. A total of 26 cases of colon adenocarcinoma with metastases to regional lymph nodes were studied. Duplex immunohistochemical detection (POD+SMA and PDGFRb+SMA) was carried out by the immunohistochemical method. In most cases, POD was absent in metastases (65.4%) and PDGFRb was present (88.5%). The POD and PDGFRb staining in the invasive edge of the tumor did not correlate with metastasis. Attention was drawn to the absence of POD and PDGFRb reactions in a blood vessel embolus, as well as to negative PDGFRb in metastasis in the presence of pronounced PDGFRb in the primary tumor in patients after neoadjuvant therapy.

In this study, the effect of epinephrine on the biofilm formation of Micrococcus luteus C01 isolated from human skin was investigated in depth for the first time. This hormone has a complex effect on biofilms in various systems. In a system with polytetrafluoroethylene (PTFE) cubes, treatment with epinephrine at a physiological concentration of 4.9 × 10-9 M increased the total amount of 72-h biofilm biomass stained with crystal violet and increased the metabolic activity of biofilms, but at higher and lower concentrations, the treatment had no significant effect. On glass fiber filters, treatment with the hormone decreased the number of colony forming units (CFUs) and changed the aggregation but did not affect the metabolic activity of biofilm cells. In glass bottom plates examined by confocal microscopy, epinephrine notably inhibited the growth of biofilms. RNA-seq analysis and RT-PCR demonstrated reproducible upregulation of genes encoding Fe-S cluster assembly factors and cyanide detoxification sulfurtransferase, whereas genes encoding the co-chaperone GroES, the LysE superfamily of lysine exporters, short-chain alcohol dehydrogenase and the potential c-di-GMP phosphotransferase were downregulated. Our results suggest that epinephrine may stimulate matrix synthesis in M. luteus biofilms, thereby increasing the activity of NAD(H) oxidoreductases. Potential c-di-GMP pathway proteins are essential in these processes.

Spleen cells of BALB/c mouse immunized with the keyhole limpet haemocyanin and thyroxine conjugate were fused with P3-X63-Ag8.653 mouse myeloma cells to produce. Four monoclonal antibodies selected by indirect ELISA and partially characterized. One antibody, 1B7, chosen for immunoassay, was produced in mouse ascites fluid, purified and analyzed; it proved to belong to the IgG1 subclass. The cross-reactivity with triiodothyronine was less than 1%, the association constant was 3 x 10(9) M-1.

Spleen cells from BALB/c mice immunised with KLH-theophylline conjugate were fused with a mouse myeloma cell line P3-X63-Ag8.653, and antibody-producing hybrids were identified by enzyme immunoassay. Three cell clones were obtained, each capable of producing a unique monoclonal antibody to theophylline. Using these monoclonal antibodies, an immunoassay system for theophylline was developed.
