RESUMEN

The optimal conditions for melanin extraction from Auricularia auricula-judae (Hei 29) fruiting bodies was determined on the basis of the extract yield of melanin, calculated by using a single-factor experiment and response surface methodology. Its antioxidant activities were also studied in vitro. Various optimal process conditions for melanin extraction were determined by using Design-Expert software: incubation temperature, 69.11°C; incubation time, 58.66 minutes; and incubation pH, 12.81. Under these conditions, the melanin yield was 2.59%. We found that the antioxidant activities of A. auricula-judae melanin in vitro were strong against DPPH radicals and superoxide anions. The rate of DPPH radical scavenging was 63.04% when the A. auricula-judae melanin concentration was 0.36 mg/mL; the rate of superoxide anion scavenging reached 39.79% when the concentration was 0.375 mg/mL. However, the antioxidant activity against hydroxyl radicals was somewhat weak; the rate of scavenging reached only 7.47% when the A. auricula-judae melanin concentration was 0.06 mg/mL.

RESUMEN

Auricularia auricula-judae is an edible and medicinal fungus ranking fourth in production among the edible fungi cultivated worldwide. White villous disease is rampant in Northeast China; it infects the fruiting bodies of A. auricula-judae by forming a white mycelial layer on its ventral side. The disease not only causes an unacceptable morphological appearance and a poor-quality product, but it also significantly reduces the yield. In this study, based on fungal morphology, ribosomal DNA internal transcribed spacer sequences, identification of species-specific primers, and the pathogenicity of the mycelia and spores, 2 fungal pathogens were isolated and identified as Fusarium equiseti and F. sporotrichioides.

BACKGROUND: A colostomy can negatively impact patients' quality of life. Recent research findings suggest that self-efficacy and psychosocial adjustment are positively associated with quality of life, but there are few research reports about the self-efficacy and psychosocial adjustment of patients with a permanent colostomy. OBJECTIVES: The objective of the study was to evaluate the effect of a 3-week Expert Patient Program (EPP) on colostomy knowledge, stoma care self-efficacy, self-management, and psychosocial adjustment in Chinese patients who had a permanent colostomy. METHODS: Eleven patients with permanent colostomy volunteered to be trained to be expert patients. Eighty-one patients with permanent colostomy were recruited and participated in the 3-week EPP courses. Questionnaires with items about knowledge on colostomy, stoma care self efficacy, self-management, and psychosocial adjustment were administered before and at 4 weeks following the EPP courses. Each EPP participant also completed a verbal interview at the end of the program. RESULTS: The EPP participants demonstrated statistically significant improvement (P < .01) in knowledge, self-efficacy, self-management, and psychosocial adjustment to colostomy (mean [SD]: 51.89 [11.45] vs 89.22 [19.90], 71.56 [5.93] vs 85.61 [14.32], 125.44 [19.27] vs 140.78 [15.34], 47.59 [9.64] vs 53.37 [10.68], respectively). Most participants gave positive ratings to the EPP. CONCLUSIONS: This study showed that the EPP was applied effectively in patients with permanent colostomy. IMPLICATIONS FOR PRACTICE: This study highlights the importance of psychosocial support for patients with permanent colostomy. It offers a model of self-help practice in colostomy patients, supported by community that can benefit more patients with permanent colostomy.

Understanding the inheritance of resistance to Fusarium ear rot is a basic prerequisite for an efficient resistance breeding in maize. In this study, 250 recombinant inbred lines (RILs) along with their resistant (BT-1) and susceptible (N6) parents were planted in Zhengzhou with three replications in 2007 and 2008. Each line was artificially inoculated using the nail-punch method. Significant genotypic variation in response to Fusarium ear rot was detected in both years. Based on a genetic map containing 207 polymorphic simple sequence repeat (SSR) markers with average genetic distances of 8.83 cM, the ear rot resistance quantitative trait loci (QTL) were analyzed by composite interval mapping with a mixed model (MCIM) across the environments. In total, four QTL were detected on chromosomes 3, 4, 5, and 6. The resistance allele at each of these four QTL was contributed by resistant parent BT-1, and accounted for 2.5-10.2% of the phenotypic variation. However, no significant epistasis interaction effect was detected after a two-dimensional genome scan. Among the four QTL, one QTL with the largest effect on chromosome 4 (bin 4.06) can be suggested to be a new locus for resistance to Fusarium ear rot, which broadens the genetic base for resistance to the disease and can be used for further genetic improvement in maize-breeding programs.

A combination of Fructus psoraleae (3.4 g/kg) and dihydroartemisinin (60 mg/(kg x d)) was used for 7 d in mice infected with Cryptosporidium parvum infection induced by oral administration of dexamethasone. The results indicated that number of Cryptosporidium parvum oocysts in feces, proportion of CD4+, CD3+ T lymphocytes in blood, serum IFN-gamma, and nitric oxide (NO) content in intestinal tissue were all higher than those of infected control group (P < 0.01). The pathogenic change in intestine of treated mice was lighter than that of infected control. It indicates that the combination of Fructus psoraleae and dihydroartemisinin is effective to mice cryptosporidiosis probably through upregulating serum IFN-gamma, proportion of CD4+ and CD3+ T cells and NO concentration in intestine.

Ordered mesoporous carbons (OMCs) with varying pore size, and microporous carbon, CFY, were synthesized using ordered mesoporous silica SBA-15 and NaY zeolite as hard templates, respectively. N(2) adsorption tests show that the synthesized OMCs possess abundant mesopores and centralized mesopore distribution. Methylene blue (MB) and neutral red (NR) were used as probe molecules to investigate their adsorption behaviors on OMCs and CFY. As evidenced by adsorption tests, the volume of mesopores of which the pore size is larger than 3.5 nm is a crucial factor for the adsorption capacity and adsorption rate of MB on OMCs. However, the most probable pore diameter of OMCs was found to be vital to the adsorption capacity and adsorption rate of NR. Theoretical studies show that the adsorption kinetics of MB and NR on OMCs can be well depicted by using pseudo-second-order kinetic model.

RESUMEN

SD rat model of PCP was established by subcutaneous injection with dexamethasone. The treatment groups received Fructus Psoralea (FP, 10.0 mg/kg), Brucea javanica (BJ, 1.2 mg/kg) and a mixture of the two Chinese herbs(FP 5mg/kg, BJ 0.6 mg/kg) respectively. By means of detecting the level of IL-2 in sera and NK cells in spleens, the effect of FP and BJ on the level of IL-2 and NK cells in rats with Pneumocystis carinii pneumonia (PCP) was observed, with SMZco treatment group (TMP 50 mg/kg, SMZ 250 mg/kg) and groups of infected and normal rats as controls. Compared with the infected group, the level of IL-2(526.1 +/- 5.5) pg/ml and NK cells (27.1% +/- 0.8%) significantly increased in the FP group (P < 0.01), followed by the FP/BJ combination group [(314.7 +/- 6.7) pg/ml, 22.9% +/- 0.9%) (P < 0.05)], and BJ group [(285.4 +/- 6.1) pg/ml, 20.7% +/- l.0%) (P < 0.05)]. Chinese herbs Fructus Psoralea and Brucea javanica show an immune regulatory action on the PCP rats.

RESUMEN

Rats with Pneumocystis carinii pneumonia (PCP) were established by hypodermic injection of dexamethasone and treated by Brucea javanica combined with Fructus Psoraleae 2 ml (Brucea javanica 0.12 mg and Fructus Psoraleae 1.0 mg) per rat per day for 7 days or 14 days. The effect of cyst-killing and the pathological change were observed under transmission electron microscope. Lung damage was alleviated or repaired, and a significant reduction of cysts was shown in the treatment group. The results show that a combination of Brucea javanica and Fructus Psoraleae played a significant restraining and killing effect on cysts, and helped repair the impairment of pneumonia.
