RESUMEN

Cardiovascular diseases (CVDs) are often associated with impaired nitric oxide (NO) bioavailability, a critical pathophysiological alteration in CVDs and an important target for therapeutic interventions. Recent studies have revealed the potential of inorganic nitrite and nitrate as sources of NO, offering promising alternatives for managing various cardiovascular conditions. It is now becoming clear that taking advantage of enzymatic pathways involved in nitrite reduction to NO is very relevant in new therapeutics. However, recent studies have shown that nitrite may be bioactivated in the acidic gastric environment, where nitrite generates NO and a variety of S-nitrosating compounds that result in increased circulating S-nitrosothiol concentrations and S-nitrosation of tissue pharmacological targets. Moreover, transnitrosation reactions may further nitrosate other targets, resulting in improved cardiovascular function in patients with CVDs. In this review, we comprehensively address the mechanisms and relevant effects of nitrate and nitrite-stimulated gastric S-nitrosothiol formation that may promote S-nitrosation of pharmacological targets in various CVDs. Recently identified interfering factors that may inhibit these mechanisms and prevent the beneficial responses to nitrate and nitrite therapy were also taken into consideration.

This study aims to estimate the COVID-19 vaccine acceptance and hesitancy among people living with HIV (PLWHA). A search for observational studies was conducted in five databases and preprinted literature. Summary estimates were pooled using a random effects model and meta-regression. Of 150 identified studies, 31 were eligible (18,550 PLWHA). The weighted prevalence of COVID-19 vaccine hesitancy overall was 29.07% among PLWHA (95%CI = 24.33-34.32; I² = 98%,) and that of vaccine acceptance was 68.66% (95%CI = 62.25-74.43; I² = 98%). Higher hesitancy prevalence was identified in low/lower-middle income countries (35.05; 95% CI = 19.38-54.78). The heterogeneity was explained by the risk of bias, region, and year of data collection. The findings conclude that the COVID-19 vaccine hesitancy rate remains high, especially in low-income countries. Evidence-informed interventions aimed at increasing COVID-19 vaccine acceptance at the national and individual levels ought to be designed to increase COVID-19 vaccine acceptance among PLWHA.

Pachira aquatica is a species used for medicinal and food purposes and has numerous phytochemicals that may have systemic toxic effects and damage to genetic material. This study aimed to evaluate acute and short-term oral toxicity, as well as genotoxic and clastogenic effects of oil extracted from P. aquatica (PASO) seeds in rats and Drosophila melanogaster. The results obtained with biochemical and hematological analyses did not show significant changes in any evaluated parameters when compared with reference values for the species used in the study. Data from the histopathological analysis corroborated results found in this study. These findings indicate low acute and short-term toxicity following oral PASO exposure in rats under the experimental conditions tested. Tests performed in rats showed that PASO did not present significant genotoxic or clastogenic effects on the cells analyzed with the three doses tested. Treatment with PASO in the offspring of HB crossing, which showed high cytochrome P450 levels, did not exhibit genotoxic activity, as demonstrated by the SMART test. These results suggest that products from the hepatic oil metabolism did not show genotoxicity under the conditions tested. Together, the results indicate that, under the experimental conditions tested, PASO is safe for repeated intake. As PASO exhibited low potential to cause harmful effects on living organisms, our study encourages further research aimed at assessing its pharmacological activity, since it is a widely consumed plant.

OBJECTIVES: To investigate the evidence on the association between ankyloglossia and obstructive sleep apnea. METHODS: An integrative literature review was carried out in the databases. Observational and interventional studies that assessed the lingual frenulum in children with sleep-disordered breathing were included. As exclusion criteria: studies in animals, in vitro, letters to the editor, expert opinions, other reviews. The selected articles were analyzed regarding the study design, sample, characterization of the lingual frenulum and sleep assessment, in addition to the main results and conclusions. RESULTS: Ninety-seven articles were identified, but only 4 met the inclusion criteria. Two retrospective studies concluded that the untreated shortened lingual frenulum at birth is associated with obstructive sleep apnea. A prospective study concluded that there was an improvement in speech and deglutition after lingual frenectomy, in addition to improved sleep. A retrospective cohort concluded that lingual frenuloplasty combined with myofunctional therapy is effective in the treatment of snoring and mouth breathing. CONCLUSION: The studies included in this review contribute to corroborate the association between ankyloglossia and obstructive sleep apnea.

Resumo Objetivo Investigar as evidências sobre a associação entre a anquiloglossia e a apneia obstrutiva do sono. Método Foi feita revisão de literatura integrativa nas bases de dados. Foram incluídos estudos observacionais e intervencionais em que foi feita a avaliação do frênulo de língua em crianças com distúrbios respiratórios do sono. Como critérios de exclusão: estudo em animais, in vitro, carta ao editor, opinião de expert, outras revisões. Os artigos selecionados foram analisados quanto ao desenho do estudo, casuística, caracterização da avaliação do frênulo lingual e do sono, além dos principais resultados e conclusões. Resultado Foram localizados 97 artigos, porém apenas 4 atenderam aos critérios de inclusão. Dois estudos retrospectivos concluiram que o frênulo lingual curto não tratado ao nascimento está associado à apneia obstrutiva do sono. Um estudo prospectivo concluiu que, após a frenectomia lingual, além da melhoria do sono, houve melhoria na fala e deglutição. Um coorte retrospectivo concluiu que a frenuloplastia lingual associada à terapia miofuncional é eficaz no tratamento do ronco e respiração oral. Conclusão Os estudos incluídos na presente revisão contribuem para corroborar a associação entre anquiloglossia e apneia obstrutiva do sono.

Serotonin (5-HT) receptors are found throughout central and peripheral nervous systems, mainly in brain regions involved in the neurobiology of anxiety and depression. 5-HT receptors are currently promising targets for discovering new drugs for treating disorders ranging from migraine to neuropsychiatric upsets, such as anxiety and depression. It is well described in the current literature that the brain expresses seven types of 5-HT receptors comprising eighteen distinct subtypes. In this article, we comprehensively reviewed 5-HT1-7 receptors. Of the eighteen 5-HT receptors known today, thirteen are G protein-coupled receptors (GPCRs) and represent targets for approximately 40% of drugs used in humans. Signaling pathways related to these receptors play a crucial role in neurodevelopment and can be modulated to develop effective therapies to treat anxiety and depression. This review presents the experimental evidence of the modulation of the "serotonergic receptosome" in the treatment of anxiety and depression, as well as demonstrating state-of-the-art research related to phytochemicals and these disorders. In addition, detailed aspects of the pharmacological mechanism of action of all currently known 5-HT receptor families were reviewed. From this review, it will be possible to direct the rational design of drugs towards new therapies that involve signaling via 5-HT receptors.

On March 11, 2020, the World Health Organization (WHO) officially declared the outbreak caused by the new coronavirus (SARS-CoV-2) a pandemic. The rapid spread of the disease surprised the scientific and medical community. Based on the latest reports, news, and scientific articles published, there is no doubt that the coronavirus has overloaded health systems globally. Practical actions against the recent emergence and rapid expansion of the SARS-CoV-2 require the development and use of tools for discovering new molecular anti-SARS-CoV-2 targets. Thus, this review presents bioinformatics and molecular modeling strategies that aim to assist in the discovery of potential anti-SARS-CoV-2 agents. Besides, we reviewed the relationship between SARS-CoV-2 and innate immunity, since understanding the structures involved in this infection can contribute to the development of new therapeutic targets. Bioinformatics is a technology that assists researchers in coping with diseases by investigating genetic sequencing and seeking structural models of potential molecular targets present in SARS-CoV2. The details provided in this review provide future points of consideration in the field of virology and medical sciences that will contribute to clarifying potential therapeutic targets for anti-SARS-CoV-2 and for understanding the molecular mechanisms responsible for the pathogenesis and virulence of SARS-CoV-2.

Fruit plants have been widely used by the population as a source of food, income and in the treatment of various diseases due to their nutritional and pharmacological properties. The aim of this study was to review information from the most current research about the phytochemical composition, biological and toxicological properties of four fruit species widely used by the world population in order to support the safe medicinal use of these species and encourage further studies on their therapeutic properties. The reviewed species are: Talisia esculenta, Brosimum gaudichaudii, Genipa americana, and Bromelia antiacantha. The review presents the botanical description of these species, their geographical distribution, forms of use in popular medicine, phytochemical studies and molecules isolated from different plant organs. The description of the pharmacological mechanism of action of secondary metabolites isolated from these species was detailed and toxicity studies related to them were reviewed. The present study demonstrates the significant concentration of phenolic compounds in these species and their anti-inflammatory, anti-tumor, photosensitizing properties, among others. Such species provide important molecules with pharmacological activity that serve as raw materials for the development of new drugs, making further studies necessary to elucidate mechanisms of action not yet understood and prove the safety for use in humans.

A educação construtivista é uma excelente alternativa para direcionar uma formação farmacêutica com as competências necessárias para atuar no âmbito da gestão do Sistema Único de Saúde (SUS). Este artigo tem como objetivo relatar a vivência do ambiente virtual como ferramenta de formação construtivista em saúde. Trata-se da experiência entre educador e educandos do curso de farmácia, com uso de metodologias ativas de ensino-aprendizagem baseadas no construtivismo. Em busca dessa construção, foi utilizado o webfólio como ferramenta de formação construtivista na disciplina práticas em saúde coletiva II: gestão. Assim, a proposta de utilização desse método é fundamentada à luz da obra de Paulo Freire em simbiose com Manoel de Barros. O processo de aprendizagem com a construção do webfólio e viagem educacional permitiu refletir sobre a realidade da nossa sociedade e a formação de agentes transformadores para gestão do SUS. Assim, os educandos foram empoderados e sensibilizados a seguir com o compromisso de reproduzir a corresponsabilidade no âmbito profissional, buscando a qualidade do atendimento com vistas à integralidade da assistência ao cuidado. A utilização dessas ferramentas foi assertiva por estimular uma formação por competências, interdisciplinaridade, interprofissionalidade, além de permitir nos maravilharmos com novas experimentações por meio de uma sociedade que muda permanentemente.


Constructivist education is an excellent alternative to direct pharmaceutical training with the necessary skills to act within the scope of the Unified Health System (SUS) management. This article reports the experience of virtual environments as a tool for constructivist education in health, namely the experience between educator and Pharmacy students with use of active teaching-learning methodologies based on constructivism. A webfólio was used as a tool for constructivist training in the discipline Practices in Collective Health II: Management. Thus, the theoretical framework is based on the work of Paulo Freire and Manoel de Barros. The learning process with the construction of the webfólio and educational trip allowed to reflect on the reality of our society and the formation of transforming agents for SUS management. Thus, the students were empowered and sensitized to follow the commitment to reproduce co-responsibility in professional settings, seeking the quality of care while aiming for the integrality of care. The use of these tools was assertive as it stimulated education based on competences, interdisciplinarity, interprofessionality, in addition to allowing us to marvel at new experiments through a society that changes permanently.


La educación constructivista es una excelente alternativa para brindar la formación farmacéutica con las competencias necesarias para actuar en el ámbito de la gestión del Sistema Único de Salud (SUS). Este artículo tiene como objetivo relatar la vivencia del ambiente virtual como una herramienta de formación constructivista en salud. Esta es la experiencia entre educador y alumnos del curso de Farmacia con el uso de metodologías activas de enseñanza-aprendizaje basadas en el constructivismo. Para esa construcción se utilizó el webfólio como herramienta de formación constructivista en la disciplina Prácticas en Salud Colectiva II: Gestión. Así la propuesta de utilización de este método está fundamentada bajo la luz de la obra de Paulo Freire en simbiosis con Manoel de Barros. La utilización de esas herramientas permitió estimular una formación por competencias, interdisciplinaridad, interprofesionalidad, además de sorprendernos con nuevas experiencias en una sociedad que cambia permanentemente.

Study based on qualitative research, from an interpretative perspective. Its objective was to understand the therapeutic itinerary of people with Diabetes Mellitus who search for different care and treatments within the different subsystems of health care. The data was collected through in-depth interviews and focus groups. As a result of the data analysis therapeutic modalities were identified, the evaluation of the care process and health treatment and the therapeutic journey in the three subsystems. The person with Diabetes Mellitus needs to reevaluate their process of living. Thus, the person circulates through various therapeutic modalities until they perceive that (or those) which are most convenient for them. This also applies to how said care or treatment becomes integrated into their day-to-day.

Estudo fundamentado na pesquisa qualitativa, na perspectiva interpretativista. Teve como objetivo conhecer o itinerário terapêutico de pessoas com diabetes mellitus na busca de cuidados e tratamentos nos diferentes subsistemas de cuidado a saúde. Os dados foram obtidos através de entrevistas em profundidade e grupos focais. A análise permitiu identificar as modalidades terapêuticas, a avaliação do cuidado e do tratamento à saúde e o percurso terapêutico nos três subsistemas. O Diabetes Mellitus requer mudanças no processo de viver. A pessoa realiza várias modalidades terapêuticas até perceber aquela ou aquelas que lhes são mais adequadas, tanto do ponto de vista do bem estar físico, quanto de como esse cuidado ou tratamento interfere em seu cotidiano.

Estudio basado en el abordaje de investigación cualitativa, en la perspectiva interpretativa. Tuvo como objetivo conocer el itinerario terapéutico de las personas con Diabetes Mellitus en la busca de la atención y tratamientos en los diferentes subsistemas de atención a la salud. Los datos fueron obtenidos a través de entrevistas en profundidad y grupos focales. Fueron identificadas las modalidades terapéuticas, la evalución del cuidado y del tratamiento y el recorrido terapéutico en los tres subsistemas. Percibimos que el mayor impacto de vivir con diabetes mellitus es que la persona necesita revisar su proceso de vivir. Así, la persona circula por varias modalidades terapéuticas hasta percibir aquellas que les sean más convenientes, tanto desde el punto de vista del bienestar físico, como de la forma en que el tratamiento se integra a su cotidiano.
