This study evaluated the median lethal concentration of silver nanoparticles and their effects in fish tambaqui Colossoma macropomum. Therefore, an acute toxicity assay was carried out in completely randomized design evaluating six different concentrations of silver nanoparticles on blood parameters of tambaqui. The silver nanoparticles were produced by chemical reduction with polyvinyl alcohol (AgNP-PVA). The lethal concentration 50% (LC50) was estimated using probit regression. The blood was collected, analyzed and the data were submitted to T-test (dying x surviving fish) and Tukey test (surviving fish). An increase in glucose, hematocrit, total plasma protein, hemoglobin, erythrocytes, leukocytes, monocytes, and neutrophils as well as reduced MCV (mean corpuscular volume) in dying fish compared to surviving fish were observed. Survived fish exposed to 187.5 µg/L showed an increase in hematocrit, MCV, and MCH and a reduction in erythrocytes, total numbers of leukocyte, thrombocyte, lymphocyte, and neutrophil. The fish exposed to concentrations below 125 µg/L, had returned the blood parameter to baselines compared to control. The estimated LC50 was 165.09 µg/L and was classified as highly toxic for the fish tambaqui. In higher concentrations, it causes an acute respiratory toxicity, but in concentrations below 125 µg/L, the fish can adapt to the stressing agent.

This study evaluated the neuroprotective effects of the Africanized bee venom (BV) and its mechanisms of action after 6-hydroxydopamine-(6-OHDA)-induced lesion in a mice model. Prior to BV treatment, mice received intrastriatal microinjections of 6-OHDA (no induced dopaminergic neuronal death) or ascorbate saline (as a control). BV was administered subcutaneously at different dosages (0.01, 0.05 or 0.1 mg·Kg-1) once every two days over a period of 3 weeks. The open field test was carried out, together with the immunohistochemical and histopathological analysis. The chemical composition of BV was also assessed, identifying the highest concentrations of apamin, phospholipase A2 and melittin. In the behavioral evaluation, the BV (0.1 mg·Kg-1) counteracted the 6-OHDA-induced decrease in crossings and rearing. 6-OHDA caused loss of dopaminergic cell bodies in the substantia nigra pars compacta and fibers in striatum (STR). Mice that received 0.01 mg·Kg-1 showed significant increase in the mean survival of dopaminergic cell bodies. Increased astrocytic infiltration occurred in the STR of 6-OHDA injected mice, differently from those of the groups treated with BV. The results suggested that Africanized BV has neuroprotective activity in an animal model of Parkinson's disease.

Infection with Mycobacterium tuberculosis (Mtb), the etiological agent of tuberculosis (TB), remains a significant global epidemic. Host resistance to Mtb depends on both adaptive and innate immunity mechanisms, including development of antigen-specific CD4 and CD8 T cells, production of inflammatory cytokines, bacterial phagocytosis and destruction within phagolysosomes, host cell apoptosis, and autophagy. A key regulatory mechanism in innate immunity is the attachment of the small protein ubiquitin to protein and lipid targets by the enzymatic activity of ubiquitin ligases. Here, we summarize the latest advances on the role of ubiquitination and ubiquitin ligases in host immunity against Mtb, with a focus on innate immunity signaling, inflammation, and antimicrobial autophagy. Understanding how ubiquitin ligases mediate immunity to Mtb, and the specific substrates of distinct ubiquitin ligases in the context of Mtb infection, could facilitate development of new host-directed antimicrobials.

Carbon nanotubes (CN) have been studied to treat spinal cord injuries because of its electrical properties and nanometric dimensions. This work aims to develop a photopolymerizable hydrogel containing CN functionalized with an anti-inflammatory molecule to be used in situ on spinal cord injuries. The CN functionalization step was done using the drug (formononetin). The nanocomposites were characterized by morphological analysis, FTIR, Raman Spectroscopy, thermal analysis and cytotoxicity assays (MTT and HET-CAM). The nanocomposites were incorporated into gelatin methacryloyl hydrogel and exposed to UV light for photopolymerization. The volume of the formulation and the UV exposition time were also analyzed. The CN characterization showed that formononetin acted as a functionalization agent. The functionalized CN showed safe characteristics and can be incorporated in photocrosslinkable formulation. The UV exposition time for the formulation photopolymerization was compatible with the cell viability and also occurred in the injury site.

This work reports the preparation of HZSM-5/SBA-15 micro-mesoporous materials with composite structure by mechanosynthesis and their use as templates to micro-mesoporous carbons, with palm oil cooking waste as carbon source. HZSM-5/SBA-15 composites were prepared in different mass proportions (25:75, 50:50 and 75:25) by a mechanosynthetic strategy. The palm oil waste was infiltrated into the template pores by a two-step procedure involving grinding followed by stirring the suspension in an oil solution and carbonization under inert atmosphere. SEM images showed the efficiency of mechanosynthesis for all mass proportions, while textural properties were strongly influenced by the presence of SBA-15. A comparison of 50% HZSM-5/SBA-15 mixture with pristine SBA-15 and uncalcined SBA-15 as carbon templates evidenced that the 50% HZSM-5/SBA-15 mixture lead to the carbon with the highest surface area and pore volume. The resulting carbon material was studied as adsorbent to different dye molecules, including molecules with different charges and sizes. Results revealed that the carbon has a significantly higher affinity for cationic dyes, that the mesopore structure favors high diffusion rates and that the surface groups have a very high affinity for cationic dye molecules.

This work aimed to study the kinetics of bifenthrin and deltamethrin residues degradation in rice grains under exposure to ozone and evaluate the effect of ozonization on rice grains marketing standards. The grains sprayed with pesticides were exposed to ozone at a concentration of 3 mg L-1 and continuous flow of 1.0 L min-1 for defined periods of up to 10 h. Residues of pesticides were extracted from grains using solid liquid extraction method with low-temperature partition analyzed by gas chromatography with electron capture detection (SLE/LTP-GC/ECD). Data from residual concentrations of bifenthrin and deltamethrin, due to the ozone exposure period, were adjusted to kinetic models of zero order, first order and second order. It was observed that ozone was effective in the degradation bifenthrin and deltamethrin residues, removing 91.9% of bifenthrin and 92.7% of deltamethrin. The kinetic model that best fitted deltamethrin and bifenthrin residues degradation data by ozone, was the one of first order. The marketing standards evaluated were the water content and yield. It was observed the ozone did not alter the rice grains water content and yield.

The objective of this work was to study the reduction in the capsaicin toxicity by encapsulation in liposomes. Capsaicin was extracted from peppers and characterized with high performance liquid chromatography (HPLC). We determined the zeta potential, polydispersivity index (PdI) and vesicle size of liposomes. Wistar rats were submitted to intravesical instillation of liposomes (LIP), capsaicin (CAP) or liposomes with capsaicin (CAPLIP). After 24 hours, bladders were removed for histological analysis. Vesicle size ranged from 68 to 105 nm with PdI smaller than 0.2 and zeta potential around -30 mV. The vesicles maintained stability over the 14-day study. The histological analysis of the CAP group showed intense inflammation in almost all bladder layers, as well as ulcer formation. Conversely, the CAPLIP group showed a smooth inflammatory reaction and hyperemia. In conclusion, the liposomes effectively protected the bladder against the irritative action of capsaicin.

A hybrid platform based on ionic liquid-based alkoxysilane functionalized mesoporous silica was applied for the synthesis of supported Pt nanoparticles with peroxidase-like catalytic activity. The positively charged groups (imidazolium) chemically bonded to the surface provide dual-functionality as ion-exchangers to the hybrid material, firstly used for the in situ synthesis of the highly dispersed Pt nanostructures and, secondly, for the immobilization of biological species aiming biosensing purposes. The peroxidase-like catalytic activity of the SiO2/Imi/Pt material was evaluated towards the H2O2-mediated oxidation of a chromogenic peroxidase substrate (TMB), allowing the colorimetric detection of H2O2. Finally, to further explore the practical application of this nanomaterial-based artificial system, glucose oxidase (GOx) was immobilized on the catalytic porous platform and a bioassay for the colorimetric determination of glucose was successfully conducted as a model system. The enzyme-like catalytic properties of the SiO2/Imi/Pt as well as its ability to immobilize and keep active biological entities on the porous structure indicate that this hybrid porous platform is potentially useful for the development of biosensing devices.

Imidazolium groups were successfully prepared and grafted on the surface of SBA-15 mesoporous silica. The ion-exchange properties of the functionalized porous solid (SBA-15/R(+)Cl(-)) toward AuCl(4)(-) anions were evaluated through an ion-exchange isotherm. The calculated values of the equilibrium constant (log ß = 4.47) and the effective ion-exchange capacity (t(Q) = 0.79 mmol g(-1)) indicate that the AuCl(4)(-) species can be loaded and strongly retained on the functionalized surface as counterions of the imidazolium groups. Subsequently, solids containing different amounts of AuCl(4)(-) ions were submitted to a chemical reduction process with NaBH(4), converting the anionic gold species into supported gold nanoparticles. The plasmon resonance bands, the X-ray diffraction patterns, and transmission electron microscopy images of the supported gold nanoparticles before and after thermal treatment at 973 K indicate that the metal nanostructures are highly dispersed and stabilized by the host environment.

Here we report the use of a thiol-functionalized silica-gel to prepare supported CdS nanocrystals by a facile one-step procedure. Upon changing the relative proportion of the matrix we obtained nanocrystals with different average sizes and size distributions, as evidenced by spectroscopic measurements as well as TEM images. Photoluminescence spectra also indicated that the main effect of the matrix is related to the size control since the spectral profiles were found to be strongly dependent on the excitation wavelength. The performance of the material in the photocatalytic degradation of two commercial dyes (methylene blue and rhodamine 6G) has been tested under sunlight radiation, showing promising results. Almost complete decolorization has been observed after 80 min of exposure, with no adsorption on the silica surface.

The three-parameter Sips adsorption model was successfully employed to modeled equilibrium adsorption data of a yellow and a red dye onto a mesoporous aminopropyl-silica, in the presence of the surfactant sodium dodecylbenzenesulfonate (DBS) from 25 to 55 degrees C. The results were evaluated in relation to the previously reported surface tension measurements. The presence of curvatures of the van()t Hoff plots suggested the presence of non-zero heat capacities terms (Delta(ads)C(p)). For the yellow dye, it is observed that the values of Delta(ads)H are almost all positive and they decrease in endothermicity, in the absence and in the presence of DBS, from 25 to 55 degrees C. For the red dye, there is an increase in endothermicity in relation to the temperature increase. The negative Delta(ads)G values indicate spontaneous adsorption processes. Almost all adsorption entropy values (Delta(ads)S) were positive. This suggests that entropy is a driving force of adsorption. The adsorption thermodynamic parameters were also evaluated using a new 2(3) full factorial design analysis. The multivariate polynomial modelings indicated that the thermodynamic parameters are also affected by important interactive effects of the experimental factors and not by the temperature changes alone.
