RESUMEN

This dataset consists of video files that were created to test the accuracy of background segmentation algorithms contained in the C++ wrapper 'BGSLibrary' v3.0.0 developed by Andrews Sobral. The comparison is based on segmentation accuracy of the algorithms on a series of indoor color-depth video clips of a single person's head and upper body, each highlighting a common factor that can influence the accuracy of foreground-background segmentation. The algorithms are run on the color image data, while the 'ground truth' is semi-automatically extracted from the depth data. The camera chosen for capturing the videos features paired color-depth image sensors, with the color sensor having specifications typical of mobile devices and webcams, which cover most of the use cases for these algorithms. The factors chosen for testing are derived from a literature review accompanying the dataset as being able to influence the efficacy of background segmentation. The assessment criteria for the results were set based on the requirements of common use cases such as gamecasting and mobile communications to allow the readers to make their own judgements on the merits of each algorithm for their own purposes.

Quartz is able to incorporate trace elements (e.g., H, Li, Al, B) depending on the formation conditions (P, T, and chemical system). Consequently, quartz can be used as a tracer for petrogenetic information of silicic plutonic bodies. In this experimental study, we provide the first data set on the OH defect incorporation in quartz from granites over a pressure/temperature range realistic for the emplacement of granitic melts in the upper crust. Piston cylinder and internally heated pressure vessel synthesis experiments were performed in a water-saturated granitic system at 1-5 kbar and 700-950 °C. Crystals from successful runs were analysed by secondary ion mass spectrometry (SIMS) and Fourier transform infrared (FTIR) spectroscopy, and their homogeneity was verified by FTIR imaging. IR absorption bands can be assigned to specific OH defects and analysed qualitatively and quantitatively and reveal that (1) the AlOH band triplet at 3310, 3378 and 3430 cm-1 is the dominating absorption feature in all spectra, (2) no simple trend of total OH defect incorporation with pressure can be observed, (3) the LiOH defect band at 3470-3480 cm-1 increases strongly in a narrow pressure interval from 4 kbar (220 µg/g H2O) to 4.5 kbar (500 µg/g H2O), and declines equally strong towards 5 kbar (180 µg/g H2O). Proton incorporation is charge balanced according to the equation H+ + A+ + P5+ = M3+ + B3+, with A+ = alkali ions and M3+ = trivalent metal ions.

BACKGROUND: Meniscal calcification is considered to play a relevant role in the pathogenesis of osteoarthritis of the knee. Little is known about the biology of acetabular labral disease and its importance in hip pathology. Here, we analyze for the first time the calcification of the acetabular labrum of the hip (ALH) and its relation to hip cartilage degeneration. METHODS: In this cross-sectional post-mortem study of an unselected sample of the general population, 170 ALH specimens and 170 femoral heads from 85 donors (38 female, 47 male; mean age 62.1 years) were analyzed by high-resolution digital contact radiography (DCR) and histological degeneration grade. The medial menisci (MM) from the same 85 donors served as an intra-individual reference for cartilage calcification (CC). Scanning electron microscopy (SEM), energy dispersive analysis (ED) and Raman spectroscopy were performed for characterization of ALH CC. RESULTS: The prevalence of CC in the ALH was 100% and that in the articular cartilage of the hip (ACH) was 96.5%. Quantitative analysis revealed that the amount of ALH CC was higher than that in the ACH (factor 3.0, p < 0.001) and in the MM (factor 1.3, p < 0.001). There was significant correlation between the amount of CC in the fibrocartilage of the left and right ALH (r = 0.70, p < 0.001). Independent of age, the amount of ALH CC correlated with histological degeneration of the ALH (Krenn score) (r = 0.55; p < 0.001) and the ACH (Osteoarthritis Research Society International (OARSI), r = 0.69; p < 0.001). Calcification of the ALH was characterized as calcium pyrophosphate dihydrate deposition. CONCLUSION: The finding that ALH fibrocartilage is a strongly calcifying tissue is unexpected and novel. The fact that ALH calcification correlates with cartilage degeneration independent of age is suggestive of an important role of ALH calcification in osteoarthritis of the hip and renders it a potential target for the prevention and treatment of hip joint degeneration.

The kinetics of CH(4)-C(2)H(6) replacement in gas hydrates has been studied by in situ neutron diffraction and Raman spectroscopy. Deuterated ethane structure type I (C(2)H(6) sI) hydrates were transformed in a closed volume into methane-ethane mixed structure type II (CH(4)-C(2)H(6) sII) hydrates at 5 MPa and various temperatures in the vicinity of 0 degrees C while followed by time-resolved neutron powder diffraction on D20 at ILL, Grenoble. The role of available surface area of the sI starting material on the formation kinetics of sII hydrates was studied. Ex situ Raman spectroscopic investigations were carried out to crosscheck the gas composition and the distribution of the gas species over the cages as a function of structure type and compared to the in situ neutron results. Raman micromapping on single hydrate grains showed compositional and structural gradients between the surface and core of the transformed hydrates. Moreover, the observed methane-ethane ratio is very far from the one expected for a formation from a constantly equilibrated gas phase. The results also prove that gas replacement in CH(4)-C(2)H(6) hydrates is a regrowth process involving the nucleation of new crystallites commencing at the surface of the parent C(2)H(6) sI hydrate with a progressively shrinking core of unreacted material. The time-resolved neutron diffraction results clearly indicate an increasing diffusion limitation of the exchange process. This diffusion limitation leads to a progressive slowing down of the exchange reaction and is likely to be responsible for the incomplete exchange of the gases.

RESUMEN

Most known methods for the determination of the structure of macromolecular complexes are limited or at least restricted at some point by their computational demands. Recent developments in information technology such as multicore, parallel and GPU processing can be used to overcome these limitations. In particular, graphics processing units (GPUs), which were originally developed for rendering real-time effects in computer games, are now ubiquitous and provide unprecedented computational power for scientific applications. Each parallel-processing paradigm alone can improve overall performance; the increased computational performance obtained by combining all paradigms, unleashing the full power of today's technology, makes certain applications feasible that were previously virtually impossible. In this article, state-of-the-art paradigms are introduced, the tools and infrastructure needed to apply these paradigms are presented and a state-of-the-art infrastructure and solution strategy for moving scientific applications to the next generation of computer hardware is outlined.

The reactivity of xenon with terrestrial oxides was investigated by in situ synchrotron x-ray diffraction. At high temperature (T > 500 kelvin), some silicon was reduced, and the pressure stability of quartz was expanded, attesting to the substitution of some xenon for silicon. When the quartz was quenched, xenon diffused out and only a few weight percent remained trapped in samples. These results show that xenon can be covalently bonded to oxygen in quartz in the lower continental crust, providing an answer to the missing xenon problem; synthesis paths of rare gas compounds are also opened.

RESUMEN

A novel method for the hydrothermal synthesis of (17)O-enriched alpha-Al(2)O(3) is presented. Its advantage compared to commonly used methods is that Al(2)O(3) is directly produced from metallic Al and (17)O-enriched H(2)O without formation of intermediate Al-hydroxide phases that require subsequent dehydration. This is a much more efficient use of the (17)O label as the Al(2)O(3) has similar (17)O enrichment level as the H(2)O initially used.

PURPOSE: Neuroborreliosis may cause various neuro-ophthalmological complications. We describe a case with a bilateral optic neuropathy. CASE REPORT: A 58-year-old female developed facial paresis six weeks after an insect bite. One week later she developed bilateral optic disc swelling with haemorrhages and nerve fibre bundle defects in the lower visual field of the left eye. In CSF and serum, raised IgM and IgG titres to Borrelia burgdorferi were found. Systemic antibiotic treatment led to improvement of the vision and facial paresis, but not all visual field defects resolved, probably due to ischemic lesions of the optic disc. DISCUSSION/CONCLUSIONS: In optic nerve lesions due to neuroborreliosis it is difficult to distinguish between inflammatory and ischemic lesions. This patient demonstrated features of an ischemic optic nerve lesion.

BACKGROUND: The preventive value of vaccinations is generally accepted. Public recommended vaccinations are administered frequently and therefore even rare complications may occur. PATIENT: We report on a 56-year-old patient who suffered from an acute unilateral optic neuritis, following ten days after vaccination against diphtheria, tetanus and poliomyelitis. A complete remission occurred within six weeks after intravenous megadose prednisolone. CONCLUSIONS: Neurological and ophthalmological complications following vaccinations are rare, and in most cases reversible.

RESUMEN

HMSN is a hereditary symmetric ascending neuropathy of the peripheral nerves. Several cases of HMSN with motility disorders of both eyes have been reported in the literature. We describe a patient with HMSN type 1 and an incomplete peripheral 3rd nerve palsy of the right eye. To the best of our knowledge, this is the first report of developed unilateral oculomotor disorder in a patient with HMSN.

To clarify further the relationships between parathyroid glands and the development of hypertension, we studied the effect of cross-transplantation of these glands from young hypertensive rats in normotensive recipients. The parathyroid glands were isolated in 5-week-old hypertensive rats of the Lyon (male and female) and Milan (only male) strains and immediately grafted into the corresponding, just parathyroidectomized normotensive rats of the same age. Control rats were either sham-operated or grafted with the glands of the same normotensive strain. Plasma calcium concentration immediately decreased after parathyroidectomy (PTX) and returned to near normal values 3 weeks after the graft. Systolic blood pressure increased slightly, but significantly, in normotensive animals grafted with hypertensive glands compared with that in normotensive control rats (mean increase, +9 mm Hg in males; +5 mm Hg in females). In conclusion, parathyroid gland transplantation from the hypertensive strain is able to chronically enhance blood pressure in the normotensive animal. The parathyroid hypertensive factor recently described may be implicated in these two hypertensive strains. Our data extend observations obtained previously in SHRs and stroke-prone SHRs and add further evidence for a major function of parathyroid glands in experimental hypertension.

To evaluate the response of an epithelial barrier to a moderate but sustained oxidative stress, we cultured monolayers of Madin Darby canine kidney cells on microporous filters and exposed them to the hypoxanthine-xanthine oxidase (HX-XO) reaction. The transepithelial permeability coefficient for mannitol (Pm) was assessed as a marker of paracellular permeability. When the oxidative stress was limited in intensity and duration (production of 10 nmol/ml/min O2- with generation of 467 +/- 30 nmol/ml H2O2 over 1 h), we observed an increase of Pm with a delay of several hours (324 +/- 65% of baseline by 6 h, P < 0.005). There was complete return to control values by 24 h. The increase of Pm did not appear to be related to a depletion of cellular ATP. Protein kinase C (PKC) activity did not increase, and the rise in Pm was not prevented by CGP 41,251, a specific inhibitor of PKC. By contrast, CGP 41,251 inhibited the Pm increase that was elicited by PDBU, a phorbol ester that activates PKC. In our model, we conclude that a reversible increase of paracellular permeability occurs after oxidative stress independently of ATP depletion or PKC activation. Other, as yet unknown mechanisms have to be involved in this process.

The intracellular distribution of enzymes involved in the Crassulacean acid metabolism (CAM) has been studied in Bryophyllum calycinum Salisb. and Crassula lycopodioides Lam. After separation of cell organelles by isopycnic centrifugation, enzymes of the Crassulacean acid metabolism were found in the following cell fractions: Phosphoenolpyruvate carboxylase in the chloroplasts; NAD-dependent malate dehydrogenase in the mitochondria and in the supernatant; NADP-dependent malate dehydrogenase and phosphoenolpyruvate carboxykinase in the chloroplasts; NADP-dependent malic enzyme in the supernatant and to a minor extent in the chloroplasts; NAD-dependent malic enzyme in the supernatant and to some degree in the mitochondria; and pyruvate; orthophosphate dikinase in the chloroplasts. The activity of the NAD-dependent malate dehydrogenase was due to three isoenzymes separated by (NH4)2SO4 gradient solubilization. These isoenzymes represented 17, 78, and 5% of the activity recovered, respectively, in the order of elution. The isoenzyme eluting first was associated with the mitochondria and the second isoenzyme was of cytosolic origin, while the intracellular location of the third isoenzyme was probably the peroxisome. Based on these findings, the metabolic path of Crassulacean acid metabolism within cells of CAM plants is discussed.

Two isoenzymes each of glucosephosphate isomerase (EC 5.3.1.9), phosphoglucomutase (EC 2.7.5.1), glucose-6-phosphate dehydrogenase (EC 1.1.1.49) and 6-phosphogluconate dehydrogenase (EC 1.1.1.43) were separated by (NH4)2SO4 gradient solubilization and DEAE-cellulose ion-exchange chromatography from green leaves of the C3-plants spinach (Spinacia oleracea L.), tobacco (Nicotiana tabacum L.) and wheat (Triticum aestivum L.), of the Crassulacean-acid-metabolism plants Crassula lycopodioides Lam., Bryophyllum calycinum Salisb. and Sedum rubrotinctum R.T. Clausen, and from the green algae Chlorella vulgaris and Chlamydomonas reinhardii. After isolation of cell organelles from spinach leaves by isopyenic centrifugation in sucrose gradients one of two isoenzymes of each of the four enzymes was found to be associated with whole chloroplasts while the other was restricted to the soluble cell fraction, implying the same intracellular distribution of these isoenzymes also in the other species.Among C4-plants, glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase were found in only one form in corn (Zea mays L.), sugar cane (Saccharum officinarum L.) and Coix lacrymajobi L., but as two isoenzymes in Atriplex spongiosa L. and Portulaca oleracea L. In corn, the two dehydrogenases were mainly associated with isolated mesophyll protoplasts while in Atriplex spongiosa they were of similar specific activity in both mesophyll protoplasts and bundle-sheath strands. In all five C4-plants three isoenzymes of glucosephosphate isomerase and phosphoglucomutase were found. In corn two were localized in the bundle-sheath strands and the third one in the mesophyll protoplasts. The amount of activity of the enzymes was similar in each of the two cell fractions. Apparently, C4 plants have isoenzymes not only in two cell compartments, but also in physiologically closely linked cell types such as mesophyll and bundle-sheath cells.

Cell organelles were isolated from the CAM plants Crassula lycopodioides Lam., Bryophyllum calycinum Salisb. and Sedum rubrotinctum R.T. Clausen by isopycnic centrifugation in sucrose gradients. The inclusion of 2.5% Ficoll in the grinding medium proved to be essential for a satisfactory separation of cell organelles during the subsequent centrifugation. Peroxisomes, mitochondria, and whole and broken chloroplasts were at least partially resolved as judged by marker-enzyme-activity profiles. The isolated peroxisomes contained activities of glycollate oxidase, catalase, hydroxypyruvate reductase, glycine aminotransferase, serine-glyoxylate aminotransferase, and aspartate aminotransferase, comparable to activities found in spinach (Spinacia oleracea L.) leaf peroxisomes. In contrast to spinach, however, only little, if any, particulate malate dehydrogenase activity could be attributed to isolated peroxisomes of the three CAM plants.

Peroxisomes, whole chloroplasts, mitochondria, and broken chloroplasts of spinach (Spinacia oleracea L.), each form 1 band at its typical density, when isolated in sucrose gradients by isopycnic centrifugation in glycylglycine buffer. In potassium-phosphate buffer peroxisomes form a 2nd band at the density of whole chloroplasts. The phosphate effect is half-saturated at a concentration of 10-20 mM. If whole chloroplasts are removed by differential centrifugation before isopycnic centrifugation no second band is formed. Arsenate can be substituted for potassium phosphate while KCl, NaCl, KNO3 and glycolate cannot, showing inorganic phosphate to be the active ion. Evidence is presented showing that during isopycnic centrifugation more slowly sedimenting peroxisomes have to move through faster sedimented bands of whole and broken chloroplasts. In the presence of inorganic phosphate this leads to a specific interaction between whole chloroplasts and peroxisomes visible as a second peroxisomal band at the density of the whole chloroplasts. The relationship of the interaction in vitro to the known association of the two organelles in vivo is considered.
