RESUMEN

The system of fertilizing agricultural soils with sludge or compost from wastewater treatment processes, as one of the principles of the circular economy, can lead to microplastic (MP) contamination. The existing technical standards for fertilization are very recent and do not consider this problem, although there is scientific evidence of their existence. Therefore, this study, on the one hand, evaluates the presence of MPs in agricultural soils, previously treated with sludge or compost from wastewater treatment plants for fertilization, and on the second hand, it studies the effect of these MPs on earthworms in three different locations in the south of Spain. For the study, selected composts deriving from the different stages of the composting process and three fertilized soils with increasing MP doses were followed. Samples were taken from different sections in depth (0-5, 5-10, and 10-20 cm) to study the shape, size, type, and abundance of MPs using infrared spectroscopy (FTIR). The results showed that the most abundant shape was fiber, followed by fragment and finally bulk, for both composts and soils. Regarding size distribution, 100 µm was the predominant size in composts (64.3% ± 9.8), while in the case of soils, the predominant range was from 100 to 500 µm. The prevalent polymers in both, composts and soils, were PTFE, TPE, PP, and PET, with four times higher amounts in composts than in soils. Ingestion of common MPs were also verified in two earthworm species, which ingested concentrations higher than 2.1% w/w. PP was the most ingested MP and Eisenia fetida was more voracious compared with Lumbricus terrestris. Therefore, it can be considered a suitable bioindicator for monitoring microplastic contamination in agricultural soil.

The increasing use of plastics and the growing concern about their impact on the environment and living beings makes it necessary to study how microplastics (MP) affect aquaculture systems. In order to gain an in-depth understanding of these systems, this study covers the water intake, the purification treatment at the inlet, the water in the culture tanks, as well as the feed used in the feeding and the organism itself. For this purpose, five samples were taken, both in the water line, feed and sea bass during the weeks of the experiment. It is shown that the available purification systems reduce the amount of MP entering from the receiving environment. However, new MP are observed in the sea bass tank, which may be due mainly to those added through the feed and found in the feed, as well as in the piping and other materials used in current aquaculture systems (PTFE, PA, among others). If focusing on the feed that can reach the consumer, in the case of this study, carried out with sea bass, some types of MP (PE, PTFE, PS and PA) were found in 4 head samples and 4 skin/muscle samples. Although inlet water purification systems manage to reduce a high percentage of MPs in the system, it is observed that there are other access routes that should be considered and reduced in aquaculture facilities to prevent them from reaching the human consumer.

Microplastic (MPs) contamination is ubiquitous in most terrestrial and aquatic ecosystems. Recently MPs have been reported at high altitudes which indicates that air masses can transport and deposit MPs in the surface snow of high mountain ecosystems, however, whether MPs typification and abundance can be influenced by direction and origin of air masses still remains an open question. Here we present the first report of MPs above 5000 m a.s.l from surface snow of a glacier in the tropical Andes. We collected surface snow along an elevational gradient, from 5000 to 5400 m a.s.l., in the Antisana Glacier, in the northern Andes cordillera of Ecuador to analyze MPs abundance and polymeric identification with the Fourier Transform Infrared (FTIR) and also to hypothesized the possible MPs sources in this remote area by comparing the oxygen and hydrogen stable isotopic ratio composition of the snow samples and by analyzing the wind direction. We observed an average of 131 ± 24 MPs L-1 in our samples. Fibers corresponded to 70% of all MP shapes; FTIR results showed that MPs composition mainly included polyurethane, polyethylene, polyamide, polyester, and high-density polyethylene in surface snow. There were no statistically significant differences of MPs abundance among sampled elevations, and the isotopic ratio composition did not differ among locations. Our results suggest that MP that accumulated in the glacier may be transported from the east, across the Amazonia, by the prevalent eastward air flow. The absence of industrial cities at least 2000 km further east from Antisana, indicates that the remote Andean glaciers could constitute important depositional zones for long-distance transported contaminants.

Pharmaceutical products, as well as insecticides and antimicrobials, have been extensively studied, but knowledge of their effects-especially those caused by their mixtures with microplastics-on aquatic organisms remains limited. However, it should be borne in mind that the state of knowledge on acute and chronic effects in aquatic organisms for pharmaceuticals and pesticides is not similar. In response, this investigation analyzed the presence of microplastics (polyvinyl chloride) and their impacts on the toxicity of chlorpyrifos (an insecticide) and triclosan (an antibacterial) when they coincide in the environment, alongside the two most consumed drugs of their type (hypolipemic and anticonvulsant, respectively), namely simvastatin and carbamazepine, in Artemia salina. LC50 and cholinesterase enzyme activity were calculated to determine the possible neurotoxicity associated with emergent contaminants in the treatments. The LC50 values obtained were 0.006 mg/dm3 for chlorpyrifos, 0.012 mg/dm3 for chlorpyrifos associated with microplastics, 4.979 mg/dm3 for triclosan, 4.957 mg/dm3 for triclosan associated with microplastics, 9.35 mg/dm3 for simvastatin, 10.29 mg/dm3 for simvastatin associated with microplastics, 43.25 mg/dm3 for carbamazepine and 46.50 mg/dm3 for carbamazepine associated with microplastics in acute exposure. These results indicate that the presence of microplastics in the medium reduces toxicity, considering the LC50 values. However, exposure to chlorpyrifos and carbamazepine, both alone and associated with microplastics, showed a decline in cholinesterase activity, confirming their neurotoxic effect. Nevertheless, no significant differences were observed with the biomarker cholinesterase between the toxicant and the toxicant with microplastics.

CbrAB is a high ranked global regulatory system exclusive of the Pseudomonads that responds to carbon limiting conditions. It has become necessary to define the particular regulon of CbrB and discriminate it from the downstream cascades through other regulatory components. We have performed in vivo binding analysis of CbrB in P. putida and determined that it directly controls the expression of at least 61 genes; 20% involved in regulatory functions, including the previously identified CrcZ and CrcY small regulatory RNAs. The remaining are porines or transporters (20%), metabolic enzymes (16%), activities related to protein translation (5%) and orfs of uncharacterised function (38%). Amongst the later, we have selected the operon PP2810-13 to make an exhaustive analysis of the CbrB binding sequences, together with those of crcZ and crcY. We describe the implication of three independent non-palindromic subsites with a variable spacing in three different targets; CrcZ, CrcY and operon PP2810-13 in the CbrAB activation. CbrB is a quite peculiar σN-dependent activator since it is barely dependent on phosphorylation for transcriptional activation. With the depiction of the precise contacts of CbrB with the DNA, the analysis of the multimerisation status and its dependence on other factors such as RpoN o IHF, we propose a model of transcriptional activation.
