RESUMEN

The article describes the phenomenon of a pharmacy in the Jewish ghetto in Terezín (Theresienstadt) in connection with the local healthcare system and the history of this ghetto. It lists the names of the Czechoslovak Jewish pharmacists who passed through this ghetto, including their fates, whether they survived or were murdered in extermination concentration camps or died as a result of the cruel living conditions in the ghetto. The article discusses the fate of the so-called Mischlingskinder ("mixed children", i.e., persons deemed to have both "Aryan" and Jewish ancestry) and "Aryan" men and women from the so-called "mixed" marriages. In a separate section, the attention is also paid to the fate of Jewish pharmacists from Germany and Austria. In all chapters, the data illustrated by the fate of some pharmacists are stated. RESUMEN

In interwar Czechoslovakia health care, an increased attention paid to the new ideas of scientific management (Taylorism), work rationalization and standardization led to the establishment of the Commission for Rationalization and Standardization in Medicine, Veterinary Medicine and Pharmacy (RANOK) within the Department of Natural Science and Medicine of the Masaryk Academy of Work. Within RANOK, the group for pharmacy worked between 1928 and 1932. The first part of the paper described the scientific management and standardization movement in interwar Czechoslovakia, the establishment of Masaryk Academy of Work and RANOK, and work objectives of RANOK and its group for pharmacy. The second part deals with the work results, relative failure and importance of the group for pharmacy.

UNLABELLED: In the 1920s Czechoslovakia, an increased attention was paid to the new ideas of scientific management (Taylorism), work rationalization and standardization. This was reflected in the foundation of the Masaryk Academy of Work in 1920. An effort to implement the new principles into health care led to the establishment of the Commission for Rationalization and Standardization in Medicine, Veterinary Medicine and Pharmacy (RANOK) within the Department of Natural Science and Medicine of the Academy. Within RANOK, the group for pharmacy worked between 1928-1932. The first part of the paper describes the scientific management and standardization movement in interwar Czechoslovakia, and the establishment of Masaryk Academy of Work and RANOK, including the group for pharmacy. The paper discusses the work objectives of the commission and presents concise biographies of the group for pharmacy members, too. The second part will be focused on the work results, relative failure and role of the group. beta(2)-Glycoprotein-I (beta(2)gpI), an abundant plasma glycoprotein, functions as a regulator of thrombosis. Previously, we demonstrated that plasmin-clipped beta(2)gpI (cbeta(2)gpI) exerts an anti-angiogenic effect on human umbilical vein endothelial cells (HUVEC). The present study was focused on the molecular background responsible for this phenomenon. cbeta(2)gpI strongly reduced HUVEC growth and proliferation as evidenced by the MTT and BrdU assay and delayed cell cycle progression arresting HUVEC in the S-and G2/M-phase. Western blot analysis indicated that cbeta(2)gpI inhibited cyclin A, B and D1, and enhanced p21 and p27 expression. Activity of p38 was down-regulated independently from the cbeta(2)gpI incubation time. Phosphorylation of ERK1/2 was not changed early (30 and 60 min) but became enhanced later (90 min, 4h). JNK activity was reduced rapidly after cbeta(2)gpI treatment but compared to controls, increased thereafter. Annexin II blockade prevented growth inhibition and cell cycle delay evoked by cbeta(2)gpI. We assume that cbeta(2)gpI's effects on HUVEC growth is mediated via cyclin A, B and D1 suppression, up-regulation of p21 and p27 and coupled to modifications of the mitogen-activated protein (MAP) kinase signalling pathway. cbeta(2)gpI may represent a potential endogenous angiogenesis-targeted compound, opening the possibility of a novel tool to treat cancer.

Six different isoquinoline alkaloids (sanguinarine, chelerythrine, berberine, coptisine, allocryptopine, and protopine) were extracted by butanol and octanol from aqueous solution, pH 4.5. The samples were analyzed by HPLC. Butanol extraction was non-selective, alkaloids passed into organic phase in 83-98%. Octanol extraction provided more selective yields: sanguinarine 99%, chelerythrine 94%, berberine 18%, coptisine 16%, allocryptopine 7.5%, protopine 7%. Further, we tested octanol treatment of extract from Dicranostigma lactucoides. The octanol extraction yields were also selective: sanguinarine 98%, chelerythrine 92%, chelirubine 92.5%, protopine 6% and allocryptopine 3.5%. 6-Butoxy-5,6-dihydrosanguinarine and 6-butoxy-5,6-dihydrochelerythrine were prepared and their NMR and MS data are reported and discussed.

The small guanine-nucleotide-binding protein Rap1 plays a key role in platelet aggregation and hemostasis, and we recently identified Rap1GAP2 as the only GTPase-activating protein of Rap1 in platelets. In search of Rap1GAP2-associated proteins, we performed yeast-2-hybrid screening and found synaptotagmin-like protein 1 (Slp1) as a new binding partner. We confirmed the interaction of Rap1GAP2 and Slp1 in transfected COS-1 and HeLa cells and at endogenous level in human platelets. Mapping studies showed that Rap1GAP2 binds through amino acids T524-K525-X-T527 within its C-terminus to the C2A domain of Slp1. Slp1 contains a Rab27-binding domain, and we demonstrate that Rap1GAP2, Slp1, and Rab27 form a trimeric complex in transfected cells and in platelets. Purified Slp1 dose-dependently decreased dense granule secretion in streptolysin-O-permeabilized platelets stimulated with calcium or guanosine 5'-O-[gamma-thio] triphosphate. The isolated C2A domain of Slp1 had a stimulatory effect on granule secretion and reversed the inhibitory effect of full-length Slp1. Purified Rap1GAP2 augmented dense granule secretion of permeabilized platelets, whereas deletion of the Slp1-binding TKXT motif abolished the effect of Rap1GAP2. We conclude that Slp1 inhibits dense granule secretion in platelets and that Rap1GAP2 modulates secretion by binding to Slp1.
