RESUMEN

Pancreatic ß­cells are the only cells that synthesize insulin to regulate blood glucose levels. Various conditions can affect the mass of pancreatic ß­cells and decrease insulin levels. Diabetes mellitus is a disease characterized by insulin resistance and chronic hyperglycemia, mainly due to the loss of pancreatic ß­cells caused by an increase in the rate of apoptosis. Additionally, hyperglycemia has a toxic effect on ß­cells. Although the precise mechanism of glucotoxicity is not fully understood, several mechanisms have been proposed. The most prominent changes are increases in reactive oxygen species, the loss of mitochondrial membrane potential and the activation of the intrinsic pathway of apoptosis due to p53. The present review analyzed the location of p53 in the cytoplasm, mitochondria and nucleus in terms of post­translational modifications, including phosphorylation, O­GlcNAcylation and poly­ADP­ribosylation, under hyperglycemic conditions. These modifications protect p53 from degradation by the proteasome and, in turn, enable it to regulate the intrinsic pathway of apoptosis through the regulation of anti­apoptotic and pro­apoptotic elements. Degradation of p53 occurs in the proteasome and depends on its ubiquitination by Mdm2. Understanding the mechanisms that activate the death of pancreatic ß­cells will allow the proposal of treatment alternatives to prevent the decrease in pancreatic ß­cells.

Overstimulation of pancreatic ß-cells can lead to dysfunction and death, prior to the clinical manifestations of type 2 diabetes (T2D). The excessive consumption of carbohydrates induces metabolic alterations that can affect the functions of the ß-cells and cause their death. We analyzed the role of p53 in pancreatic ß cell death in carbohydrate-supplemented Sprague Dawley rats. For four months, the animals received drinking water containing either 40% sucrose or 40% fructose. The glucose tolerance test was performed at week 15. Apoptosis was assessed with the TUNEL assay (TdT-mediated dUTP-nick end-labeling). Bax, p53, and insulin were assessed by Western blotting, immunofluorescence, and real-time quantitative PCR. Insulin, triacylglycerol, and serum glucose and fatty acids in pancreatic tissue were measured. Carbohydrate consumption promotes apoptosis and mobilization of p53 from the cytosol to rat pancreatic ß-cell mitochondria before blood glucose rises. An increase in p53, miR-34a, and Bax mRNA was also detected (P < 0.001) in the sucrose group. As well as hypertriglyceridemia, hyperinsulinemia, glucose intolerance, insulin resistance, visceral fat accumulation, and increased pancreatic fatty acids in the sucrose group. Carbohydrate consumption increases p53 and its mobilization into ß-cell mitochondria and coincides with the increased rate of apoptosis, which occurs before serum glucose levels rise.

The aim of this study was to determine the potential fertilizing of spermatozoa from the epididymal tail in different periods of time post-orchiectomy (P-OQ). Therefore, the study was approached in two stages. In the first stage, the orchiectomy was performed in 30 adult pigs. The testicles were stored at 5.00 ˚C in physiological saline solution for 5, 24, 48, 72, 96 and 120 hr. The spermatozoa were obtained by backflushing the vas deferens. The spermogram and fluorometric study were performed for each sample to evaluate the exposure of phosphatidyl-serine (PS) and acrosome reaction (AR). The second stage included the fertilization test, 16 prepubertal sows were selected, after synchronizing the oestrous cycle and the post-cervical artificial insemination was performed with the refrigerated sperm samples from each P-OQ time. The percentage of live sperm remained without significant changes until 96 hr P-OQ. An increase in the percentage of spermatozoa that showed a PS exposure was observed. The premature AR was evident after 72 hr. Considering that the artificial insemination was performed ensuring a minimum number of live sperms, no significant differences were observed in the number of embryos and corpora lutea. The results indicated that pig sperm collected from the epididymal tail P-OQ and stored for 5 and up to 72 hr at 5.00 ˚C had viable characteristics and maintained their fertilization ability. However, there was an increase in the loss of phospholipid asymmetry of the plasma membrane as time increased (72 and 96 hr), therefore, sperm viability was decreased.

Twenty-eight adult budgerigars were randomly divided in two groups: control (n=14) and treated (n=14) with a unique dose of synthetic GnRH-I. Testicular development was assessed through ultrasonography and density was evaluated with pixels. Germinal diameter and thickness of germinal epithelium were determined with histology; there were identified and countified different cellular strains in seminiferous tubules therefore spermatobioscopy. Results. Ecographic density was: control group: 76 ± 7 pixels, treated group 41 ± 3 pixels. Thickness of germinal epitellium, 51.5 ± 2.9µm and 73.1 ± 3.1µm, for control group and treated group respectively. Sperm concentration in the treated group was 300% superior than in control group. It is concluded that the administration of synthetic GnRH-I, is a viable alternative to be used as part of the assisted reproductive techniques to induce reproduction.

It is important to know the morphological changes that occur in the spermatozoa of rooster during their passage through the reproductive tract, which help to understand what they acquire their fertilization capacity. The morphophysiological changes related to the capacitation and acrosomal reaction processes in the different segments of the rooster reproductive system were analyzed. Samples were obtained from various regions of the rooster reproductive tract and dorso-ventral massage to obtain ejaculates, 25 roosters were used Rhode Island Red with proven fertility, assessments were performed with chlortetracycline and Lectin WGA-FITC to determine the morphophysiological parameters. Sperm motility increases (p<0.05) during the passage of spermatozoa from the testis until they are ejaculated. The parameters of viability and morphology also show differences (p<0.05) in the different segments of the tract. Sperm morphometry shows a spermatic contraction (p<0.05) in the cranial and medial segments of the vas deferens. The acrosomal reaction capacity evaluated with chlortetracycline (CTC) or Wheat germ agglutinin (WGA), was evident increasing the parameters (p<0.05) with the use of the perivitelline layer in the spermatozoa of the reproductive tract and of the ejaculate. Spermatozoa of the reproductive tract of the rooster demonstrate acrosomal reaction capacity without requiring a previous sperm capacitation condition. On the other hand, they do not show parameters of incapacity, which implies that they cannot be stored in any segment of the reproductive tract.


Es importante conocer los cambios morfológicos que se producen en los espermatozoides del gallo durante su paso por el tracto reproductivo y que ayudan a comprender como adquieren su capacidad de fertilización. Se analizaron cambios morfofisiológicos relacionados con los procesos de capacitación y reacción acrosomal de los espermatozoides presentes en los diferentes segmentos del tracto reproductor del gallo. Se obtuvieron espermatozoides de diferentes regiones del tracto reproductor del gallo y de espermatozoides de eyaculado. Se usaron 25 gallos Rhode Island Red con fertilidad probada. Se realizaron evaluaciones básicas, con clortetraciclina (CTC) y lectina Wheat germ agglutinin conjugada con isotiosionato de fluoresceína (WGA-FITC) para determinar los parámetros morfofisiológicos. La motilidad del esperma aumenta (P<0,05) durante el paso de los espermatozoides desde el testículo hasta que son eyaculados. Los parámetros de viabilidad y morfología también muestran diferencias (P <0,05) en los diferentes segmentos del tracto. La morfometría mostró una contracción de los espermatozoides (P<0,05) en los segmentos craneal y medial del conducto deferente. La capacidad de reacción acrosomal evaluada con clortetraciclina CTC o WGAFITC, fue evidente al aumentar los parámetros (P<0,05) con el uso de membrana perivitelina en los espermatozoides del tracto reproductivo y del eyaculado. los espermatozoides del tracto reproductivo del gallo demuestran capacidad de reacción acrosomal sin requerir una condición previa de capacitación espermática. Por otro lado, no muestran parámetros de descapacitación espermática lo que implica que no pueden almacenar en ningún segmento del tracto reproductivo.

The production of ornamental fishes represents an economic activity of a growing number of Mexican families. Nevertheless, the reproduction of fish in captivity is one of the complications faced by farmers. This study was set up to: (i) evaluate the morphological and functional changes induced by hydration in the gametes of fish tiger barb (Puntius tetrazona; 240 samples) at tree times after hydration (10, 20 and 30s) with classic spermograms (volume, sperm concentration, viability, motility, and normal morphology); and (ii) evaluate the implementation of in vitro fertilization based on the ovulation rate, the percentage of fertilization and hatching, and the larval numbers obtained after 72 hours. The average volume of milt was 3.0±0.7μL, and the minimum, maximum and average concentration of sperm was 44.4x10(6) spz/mL, 52.3x10(6)spz/mL, and 48.1±5.9x10(6)spz/mL, respectively. The viability and motility of the sperm was 84.6±3.2% and 81.5±2.2%, respectively. The diameter of the sperm with/without water contact was 2.1±0.6μm and 3.8±1.0μm (p<0.05); the largest diameter was recorded 30 seconds after the contact with water. For oocytes, the smaller and larger diameters were recorded at 10 and 30s, respectively (both with/without water contact); the oocytes diameters after 10 and 30 seconds of contact with water were 1.11 and 1.55mm, respectively. A higher ovulation rate was recorded using the in vitro fertilization: 250±50 oocytes versus 28±09 oocytes (during natural fertilization; p<0.05). Nevertheless, fertilization and hatching rates were higher for the natural fertilization (80 and 60%, respectively). Considering the number of larvae obtained after 72 hours, our results showed a higher value for the in vitro fertilization (75±18 compared to 13.4±12 of the natural fertilization; p<0.05). We propose this fish as a model for other ornamental fishes of commercial interest. Our results demonstrate that the in vitro fertilization is a very high viable option to optimize and maximize resources; besides, the reproduction management optimization under controlled conditions may enhance wild fish stocks preservation. El objetivo del presente estudio fue conocer las características de los gametos de Puntius tetrazona (n=240), los cambios morfológicos a partir de su activación mediante espermogramas cásicos y por otro lado, se evaluó la implementación de la fertilización in vitro a partir de la tasa ovulatoria, el % de fertilización y eclosión y el número de larvas vivas a las 72h. El volumen promedio de semen fue de 3.0±0.7µL. La concentración espermática mínima, máxima y promedio fue 44.48x10(6)spz/mL, 52.3x10(6)spz/mL y 48.1±5.9x10(6)spz/mL, respectivamente. La viabilidad promedio fue de 84.68±3.27%. La motilidad promedio fue 81.53±2.28%. El diámetro de los espermatozoides fluctuó entre 2.16±0.2 y 2.79±0.3µm; 3.84±0.3 y 4.86±0.31µm sin y con contacto con el agua respectivamente, con diferencias significativas. El diámetro mayor fue a los 30s en contacto con el agua. Los ovocitos de menor y mayor diámetro se registraron a los diez y 30s sin y con contacto con el agua respectivamente. Los diámetros de los ovocitos en diez y 30s en contacto con el agua fluctuaron entre 1.11 y 1.55mm respectivamente. La mayor tasa ovulatoria fue en la fertilización in vitro con 250±50 ovocitos frente a 28±09 de la natural, con diferencias significativas. Los porcentajes de fertilización y eclosión fueron más elevados en la fertilización natural con 80% y 60% respectivamente. Se registraron 75±18 larvas a las 72 horas en el grupo in vitro comparado con 13.4±12 larvas de la fertilización natural. Con lo anterior, la técnica que permitió mayor cantidad de larvas fue la de fertilización in vitro.

Most of the neotropical bats reproduce in a seasonal fashion. The purpose of this study was to assess changes in testicular tissue by morphometry, in Tadarida brasiliensis bat, in a south urban zone of Mexico City during summer, autumn, and winter. Three sample collections (February, June, and September) from T. brasiliensis were carried out (n=6). Testicle fragments were obtained for histological studies. Diameter of the seminiferous tubules and interstitial space were measured, cellular populations were identified and counted. June samples showed smaller diameter of seminiferous tubules and larger interstitial space between tubules; also there were less number of germinal epithelium cells and spermatids were absent. Tissues from September and February showed a significant increase (p 0.05) in tubule diameter, germinal epithelium thickness, and number of germinal epithelium cells when compared to June samples. Only February samples showed presence of spermatids. Our results suggest the existence of seasonal variations in the reproductive activity of T. brasiliensis, under conditions in which the study was conducted.


La mayoría de los murciélagos neotropicales se reproducen de manera estacional. El objetivo de este estudio fue determinar los cambios morfométricos en el parénquima testicular del murciélago Tadarida brasiliensis que habita en un área urbana del sur de la Ciudad de México durante las estaciones de verano, otoño e invierno. Se obtuvieron fragmentos de tejido testicular para su estudio histológico. Se midió el diámetro de los túbulos seminíferos y el espacio interstisial y se identificaron y contaron distintos tipos celulares. En las muestras de junio se encontró un menor diámetro de los túbulos seminíferos y un mayor espacio intersticial entre los túbulos; también hubo un menor número de células del epitelio germinal y no hubo presencia de espermátides. En las muestras de tejido obtenidas en el mes de septiembre y febrero se observó un incremento significativo en el diámetro del túbulo, grosor del epitelio germinal y número de células del epitelio germinal cuando fueron comparadas con las muestras de junio (p 0.05). Solo en las muestras de febrero hubo presencia de espermátides. En conjunto, nuestros resultados sugieren la existencia de variaciones estacionales en la actividad reproductiva de T. brasiliensis, bajo las condiciones en que se realizó el estudio.

Pancreatic ß-cell death in type 2 diabetes has been related to p53 subcellular localisation and phosphorylation. However, the mechanisms by which p53 is phosphorylated and its activation in response to oxidative stress remain poorly understood. Therefore, the aim of this study was to investigate mitochondrial p53 phosphorylation, its subcellular localisation and its relationship with apoptotic induction in RINm5F cells cultured under high glucose conditions. Our results show that p53 phosphorylation in the mitochondrial fraction was greater at ser392 than at ser15. This increased phosphorylation correlated with an increase in reactive oxygen species, a decrease in the Bcl-2/Bax ratio, a release of cytochrome c and an increase in the rate of apoptosis. We also observed a decline in ERK 1/2 phosphorylation over time, which is an indicator of cell proliferation. To identify the kinase responsible for phosphorylating p53, p38 mitogen-activated protein kinase (MAPK) activation was analysed. We found that high glucose induced an increase in p38 MAPK phosphorylation in the mitochondria after 24-72 h. Moreover, the phosphorylation of p53 (ser392) by p38 MAPK in mitochondria was confirmed by colocalisation studies with confocal microscopy. The addition of a specific p38 MAPK inhibitor (SB203580) to the culture medium during high glucose treatment blocked p53 mobilisation to the mitochondria and phosphorylation; thus, the release of cytochrome c and the apoptosis rate in RINm5F cells decreased. These results suggest that mitochondrial p53 phosphorylation by p38 MAPK plays an important role in RINm5F cell death under high glucose conditions.

Under oxidative stress, GSH regeneration is dependent on the adequate supply of NADPH by glucose-6-phosphate dehydrogenase (G6PD). In this study, we analyzed the changes of G6PD, GSH, and oxidative stress of granulosa cells and follicular liquid and its association with apoptosis during atresia of small (4-6 mm) and large (>6 mm) sheep antral follicles. G6PD activity was found to be higher in granulosa cells of healthy small rather than large follicles, with similar GSH concentration in both cases. During atresia, increased apoptosis and protein oxidation, as well as a drop in GSH levels, were observed in follicles of both sizes. Furthermore, the activity of G6PD decreased in atretic small follicles, but not in large ones. GSH decreased and protein oxidation increased in follicular fluid. This was dependent on the degree of atresia, whereas the changes in G6PD activity were based on the type of follicle. The higher G6PD activity in the small follicles could be related to granulosa cell proliferation, follicular growth, and a lower sensitivity to oxidative stress when compared with large follicles. The results also indicate that GSH concentration in atretic follicles depends on other factors in addition to G6PD, such as de novo synthesis or activity of other NADPH-producing enzymes. Finally, lower G6PD activity in large follicles indicating a higher susceptibility to oxidative stress associated to apoptosis progression in follicle atresia.

RESUMEN

En este trabajo, revisaremos las características morfológicas y las fases en que se ha dividido a la apoptosis, así como la importancia de su presencia en algunas enfermedades.La apoptosis y la necrosis son dos mecanismos mediante los cuales puede ocurrir muerte celular. La apoptosis constituye una medida fisiológica de remoción celular, bajo control genético, que se caracteriza por colapso celular, condensación de la cromatina y fragmentación del ácido desoxirribonucleico (ADN). Las células apoptóticas son rápidamente fagocitadas por células vecinas o macrófagos, previniendo así una reacción inflamatoria. La apoptosis se ha propuesto como un evento crítico para mantener la homeostasis tisular que asegura el estado de salud de los organismos.La necrosis implica la ruptura de la membrana e hipoxia, lo que conduce a la disminución en las concentraciones de adenosin trifosfato (ATP), colapso metabólico, edematización y disolución de la célula originando un proceso inflamatorio.
