RESUMEN

An immune sensor based on the surface plasmon resonance (SPR) was developed for express diagnostics of bovine leucosis. The sensor was used for detection of the level of antibodies against bovine leukaemia virus (BLV) in the blood serum. The industrially manufactured BLV antigen for screening test in the agar gel immunodiffusion (AGID) required the additional purification in order to be used in immune sensor analysis. It was shown that immune sensor analysis was more sensitive, rapid and simple in comparison with the traditional AGID test. It was stated that the developed immune sensor was capable to be used for performance of bovine leucosis screening at the farms and the minimal dilution of the serum should be 1:500.

RESUMEN

The approaches to the problems of primary diagnostics of the hereditary hematoglobinopathias caused by unbalanced synthesis of alpha and beta chains, as well as modern state of screening of hereditary thalassemic hematoglobinopathias are considered. The result of the available information analysis takes into account numerous peculiarities of this problem, including economic ones. Experience of an estimation of the displays of unbalanced synthesis of globin chains concerning the morphology of red blood cells is presented in the generalized form. The latter is a very important for preliminary diagnostics the hematoglobinopathias because, for the first, it may be realized at primary units of public health system service, for the second, it allows to narrow to an acceptable level the group of persons whose blood requires more complex and expensive researches.

RESUMEN

Primary and secondary humoral immune response (the level of specific IgM and IgG) as well as the intensity of delayed-type hypersensitivity to sheep erythrocytes were investigated in rabbits fed with apis flower pollen load for a month. It is shown that apis flower pollen load is an immunomodulator. It stimulated humoral immune response and changed the reaction of delayed-type hypersensitivity.

RESUMEN

It is found that nitrite-ions formed as a result of biotransformation during long term feeding of calves with sodium and potassium nitrates induce changes in some biochemical parameters of blood, including HS-glutathione content in erythrocytes, acid hemolytic resistance of erythrocytes, activity of NAD-dependent methemoglobin-reductase, correlation of ligand forms of hemoglobin and its total content. It is supposed that the observed changes are of an adaptational character and, as a whole, provide for the optimization of both quantitative and qualitative composition of population of erythroid cells at the expense of erythropoiesis intensification.

RESUMEN

Mildronat potentiates the therapeutic effect of the combined treatment of heart failure, which includes the use of nitro drugs. It also decreases the level of methemoglobin in the patients' blood and improves the phosphate balance, especially that of 2,3-diphosphoglycerate to control oxygen transport by hemoglobin, which is of paramount importance in hypoxia caused by coronary heart disease.

RESUMEN

The functional properties of total hemoglobin and some of its major fractions obtained by polyacrylamide gel disk-electrophoresis were studied. It was shown that p50 for intact hemoglobin is 1.18, that for fractions 2, 4-6-1.37-1.65 mm Hg. The affinity of fraction 3 for oxygen is significantly higher. In the presence of CO2 (40 mm Hg) p50 is much higher for fractions 4 and 6 as compared with fractions 3 and 5. Fraction 3 reveals the highest resistance to autooxidation; fractions 2 and 5 possess the highest sensitivity. ATP and pyridoxal-5'-phosphate stimulate the autooxidation and decrease the affinity for hemoglobin oxygen of fractions 3 and 4 in a greater degree than of the other fractions (for fraction 3 more than for fraction 4). A similar dependence is observed in case of fractions 5 and 6. The effective constant (ki) of dissociation of heme- and globin bonds for HbCO for fraction 3 is 2.38.10(-3) s-1. For the other fractions this value is considerably lower. The ki value for all fractions is increased with a transition from the carboxy- to methehemoglobin. The level of individual components of the heterogenous system of hemoglobin in the blood of embryos, adult intact rats and in anemic and hypoxic animals is given. The interrelationship of functional and structural properties of the hemoglobin fractions and their role in oxygen supply of tissues under the above-described conditions are discussed.

RESUMEN

Rat hemoglobin was fractionated by different methods. It is shown that the heterogenic system of hemoglobin includes six main fractions, the appearance of which is determined by 2 alpha- and 3 beta-similar chains. A subunit composition is established for hemoglobins certain fractions and their tetrameric structure is as follows: formula: (see text). A greater number of fractions when separating hemoglobin by high-sensitive methods is induced by its conformational changes. The surface of SH-groups of globin and the state of hem iron are found to be essentially significant in this process. The data are presented characterizing the isoelectric points, quantitative content of the fraction as well as of certain chains in the whole hemoglobin. A problem on the nature of the isolated subunits heterogeneities is under discussion.

RESUMEN

Eight fractions of rat hemoglobin obtained by electrophoresis in polyacrylamide gel were studied. Having used parachloromercuric benzoate (PChMB) and 5,5-dithio-bis (2-nitrobenzoic acid) it was established that fractions of 3 and 3a carboxyhemoglobin contain 6 available SH-groups and the rest one (1, 2, 2a, 4, 5, 6) about 4 SH-groups. In all fractions only 2 masked SH-groups are found. A problem is discussed on SH-groups localization in the polypeptide chains of certain hemoglobin fractions in rats.
