RESUMEN

Glioblastoma, which is highly invasive and has a poor patient prognosis, is the most common type of brain tumor. Flavonoids have known antiproliferative and antineoplastic effects, such as apoptosis induction and tumor growth inhibition. We investigated the effects of treatment with three flavonoids (BAS-1, BAS-4, and BAS-6) isolated from the Amazon plant Brosimum acutifolium on the proliferation and migration of the C6 glioma cell line. Cytotoxicity was evaluated by MTT assay, and morphological changes were evaluated by phase-contrast microscopy and by transmission electron microscopy. Apoptosis was determined using Annexin V-FITC-propidium iodide (PI) staining. A hemolysis assay was used to evaluate plasma membrane injury. Antiproliferative effects were assessed by wound migration and colony formation assays. Mitochondrial transmembrane potential (ΔΨm) was determined using JC-1 dye and flow cytometry. To identify the flavonoid targets, western blotting was performed. BAS-1 and BAS-4 reduced C6 cell proliferation in a dose-dependent manner. BAS-6 showed no effect. Due to its high toxicity toward primary glial cells and its high hemolytic index, BAS-1 was not used in the remaining experiments. BAS-4 treatment did not induce cytotoxicity in primary glial cells; however, in glioma cells, it suppressed migration and invasion and led to apoptosis through mitochondrial damage, ΔΨm loss, cell cycle arrest, and reduced AKT phosphorylation, which is a component of the main cell survival pathway. We conclude that BAS-4 showed potential activity against glioma by inducing apoptosis mediated by ΔΨm loss and AKT pathway disruption, and future studies should further evaluate BAS-4 as a promising antineoplastic agent against glioblastoma.

The photofragmentation dynamics of 1,1,1,2-tetrafluoroethane (R134a) with photon energies from 12 eV up to 320 eV, surrounding the C 1s edge is discussed. The ionic moieties were measured in coincidence with the ejected electrons (PEPICO mode), and detected as a function of the photon energy. Around the C K core edge, the fragmentation profiles are examined regarding the site specific excitation of the CH2FCF3 molecule. In the present case, site-selectivity is favored by the distinct chemical environments surrounding both C atoms. NEXAFS spectrum at the C 1s edge simulation has been obtained at the TDDFT level and excited state geometry optimization calculations have been performed at the inner-shell multiconfigurational self-consistent field level. Our observations indicate that the C(H2F) 1s excitation to a highly repulsive potential expels a fluorine atom leaving the heavier radical fragment C2F3H2* which relaxes to the fundamental state of the ion C2F3H2+. On the other hand, the excitation from the C(F3) 1s carbon to a repulsive state in the C-C bond, leads to a C-C bond cleavage, explaining the observed site specific fragmentation.

Solid-state compounds of the general formulae [ML3] (M=Gd, Tb, Dy, Ho, Er, Tm, Yb, Lu, Y; L=ketoprofen) were synthesized and characterized using infrared, diffuse reflectance and luminescence spectroscopies. IR data suggested that the carboxylate group in ketoprofen is coordinated to the metals as a bidentate ligand. The triplet state energy level was determined using the Gd(3+) complex, which exhibited a ketoprofen blue luminescence when excited in the UV region. The compound containing Tb(3+) ion was sensitized by the ligand and emitted in the green region of the visible spectrum. On the other hand, for the analogous species containing the dysprosium ion, a competition for luminescence between the Dy(3+) and the ligand levels was observed. Finally, Tm(3+) complex exhibits only ligand luminescence. These optical behaviors are discussed based on rare earth energy diagrams. In addition, the compounds were evaluated for their anti-inflammatory activities. All the compounds showed a higher production of H2O2 and IL-10 than the ketoprofen, suggesting that the compounds exhibited an immunomodulatory effect and this opens up new perspectives for immunotherapeutic approaches.

O objetivo deste trabalho foi realizar a caracterização físico-química do pó e da tintura, e análise por espectrofotometria e cromatografia do extrato seco de Peperomia pellucida L. (H. B. K.). As metodologias seguiram a Farmacopéia Brasileira IV ed., com exceção da prospecção química, da espectrofotometria, da obtenção do perfil cromatográfico do extrato seco, e determinação do resíduo seco. A prospecção química revelou a presença de saponinas espumídicas; açúcares redutores; proteínas e aminoácidos; fenóis; taninos; flavonóides; esteróides e triterpenóides. Na análise por CCD, o melhor perfil da fração flavonoídica foi obtido com MeOH/CHOOH (90:10). Foi confirmada, através de CLAE, a presença de 3',4',7-tri-O-metoxiflavona no extrato seco deste material vegetal. Os resultados obtidos contribuem para a determinação de especificações de uma futura monografia em Farmacopéias da Peperomia pellucida L. (H.B.K.).


The aim of this study was the physical chemical characterization of the powder and the tincture, and the chromatographic and spectrophotometric analysis of the Peperomiapellucida L. (H. B. K.) dry extract. The methodology followed the Farmacopeia Brasileira IV ed., except for the chemical prospection, the chromatographic profile obtained and the spectrophotometry of the dry extract, and determination of dried residues. The chemical prospection revealed the presence of foaming saponins; reducing sugars; proteins and amino acids; phenols; tannins; flavonoids; steroids and triterpenoids; depsideos and depsidones. The best profile from TLC for flavonoidic fraction was obtained with methanol/formic acid (90:10 v/v). HPLC confirmed the presence of 3 ',4',7-tri-methoxyflavone in the dry extract of the plant material. The results obtained in this work should contribute for the determination of specifications for a future monograph on Peperomia pellucida L. (H.B.K.).

The present study was designed to evaluate whether chronic exposure to a volcanically active environment might result in genotoxic and cytotoxic effects in human oral epithelial cells. A study group of 120 individuals inhabiting a volcanically active environment (exposed group; Furnas village) and a reference group of 122 individuals inhabiting a village without manifestations of volcanic activity (Santo António village) were examined in this study. Individuals from Furnas village inhabit a volcanically active environment marked by several degassing manifestations, including fumarolic fields, thermal and cold CO2 springs and soil diffuse degassing areas. For each individual, 1000 buccal epithelial cells were analyzed for the frequency of micronucleated cells (MNC) and the frequency of cells with other nuclear anomalies (ONA: pyknosis, karyolysis and karyorrhexis), by using the micronucleus assay. Information on life-style factors and an informed consent were obtained from each participant. The frequencies of MNC and of ONA per 1000 cells in the exposed group (4.3‰ and 23.5‰, respectively) were significantly higher than in the reference group (1.7‰ and 7.7‰, respectively). The risk of having a high frequency of MNC or ONA was 2.4 and 3.1-fold higher in exposed individuals compared to reference group. The analyzed confounding factors (age, gender, smoking status, alcohol consumption, and mouthwash use) did not show any significant association with the frequency of MNC or ONA. The higher risk of DNA damage in individuals inhabiting a volcanically active environment is for the first time clearly highlighted with this biomonitoring study. Given that MNC in oral epithelia are recognized as a predictive biomarker of cancer risk within a population of healthy subjects, these findings could contribute to explain the high incidence rates of lip, oral cavity and pharynx cancers previously referred for Furnas village inhabitants.

Óleo de Copaifera multijuga Hayne, in natura e as frações foram avaliados quanto às suas atividades fungitóxicas, frente a cinco espécies de fungos filamentosos do gênero Aspergillus e três espécies de leveduras do gênero Candida. Concentrações de óleo resina e de óleo essencial na faixa de 0,08 mg mL-1a 1,6 mg mL-1 foram usadas para as análises qualitativa e quantitativas. As amostras foram dispostas sobre discos de papel de 5 mm de diâmetro e distribuídos sobre o meio Saboraud em placas de Petri, inoculadas com esporos dos microrganismos e incubadas a 28ºC durante 10 dias. Utilizou-se uma solução com 1,6 mg mL-1 de nitrato de miconazol como controle positivo. Os resultados qualitativos mostraram que o óleo resina apresentou boa atividade, porém uma das frações do óleo essencial se mostrou altamente efetivo contra C. parapsilosis IOC - 2882, A.flavus IOC-3874 e A. tamarii IOC-187 com halos de inibição de 16,0±1,4 mm, 19,5±2,1 mm e 12,5±3,5 mm, respectivamente. Já a avaliação quantitativa mostrou que 0,3 mg mL-1 do óleo resina inibiu o crescimento de A. flavus e C. parapsilosis, enquanto que 0,08 mg mL-1 da fração do óleo essencial atingiu esta mesma atividade.


Copaifera multijuga Hayne oil in natura and its fractions were evaluated as to their fungitoxic activities against five filamentous fungus species belonging to the genus Aspergillus and three yeast species of the genus Candida. Oleoresin and essential oil concentrations ranging from 0.08 mg mL-1 to 1.6 mg mL-1 were used for qualitative and quantitative analyses. The samples were placed onto 5mm-diameter paper discs distributed on Sabouraud's medium in Petri dishes inoculated with spores of the microorganisms and incubated at 28ºC for 10 days. A solution containing 1.6 mg mL-1 miconazole nitrate was used as positive control. Qualitative results showed that the oleoresin presented good activity, while a fraction of the essential oil was highly effective against Candida parapsilosis IOC-2882, Aspergillus flavus IOC-3874 and A tamarii IOC-187 with 16.0±1.4 mm, 19.5±2.1 mm and 12.5±3.5 mm inhibition halos, respectively. The quantitative evaluation showed that 0.3 mg mL-1 oleoresin inhibited the growth of Aspergillus flavus and Candida parapsilosis, while 0.08 mg mL-1 of the essential oil fraction reached this same activity.

To better understand the interactions between opportunistic fungi and their hosts, we investigated hydrogen peroxide (H2O2), nitric oxide and TNF-alpha production by peritoneal macrophages from Ehrlich tumour-bearing mice (TBM) during microbial infections. For this purpose, TBM at days 7, 14 and 21 of tumour progression were inoculated intraperitoneally with C. albicans and evaluated after 24 and 72 h. We observed that TBM showed significant increases in H2O2, TNF-alpha levels and fungal clearance at day 7 after C. albicans infection. However, as the tumour advanced, there was a progressive decline in the release of H2O2 and TNF-alpha that was paired with the dissemination of C. albicans. These results demonstrate that protective macrophage activities against Candida albicans are limited to the initial stages of tumour growth; continued solid tumour growth weakened the macrophage response and as a consequence, weakened the host's susceptibility to opportunistic infections.

The participation of dermatophytic antigens in the host-parasite balance is still poorly understood. One of the difficulties encountered by researchers is the lack of dominant and specific antigens that can be used in such studies. In order to contribute to a better understanding of this aspect of infection, the present study identifies antigen fractions obtained from exoantigen and cytoplasmic extracts of Trichophyton mentagrophytes. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) revealed the presence of 13 proteins in the exoantigen extract, whose molecular weight ranged from 12.5 to 90 kDa. The cytoplasmic extract contained 18 protein fractions ranging from 11 to 110 kDa. Immunoblotting showed the presence of immunodominant antigens against IgG, IgM and IgA antibodies. This affinity was observed in three proteins of the exoantigen extract and in three proteins of the cytoplasmic extract, with respective molecular weights of 33, 39 and 59, and 40, 55 and 82 kDa. These results are promising, especially when considering that these extracts contain antigenically distinct protein fractions which, once determined, may contribute to a better understanding of dermatophytoses, and may thus help in the development of alternative strategies for the diagnosis and treatment of this condition.

BACKGROUND: Women with endometriosis may have higher rates of autoimmune disorders, including hypothyroidism. The objective of this study was to compare the prevalence of thyroid dysfunction and autoimmune thyroid disease (AITD) between women with endometriosis and a control group. METHODS: This was a cross-sectional study carried out in 148 women with surgically confirmed endometriosis and 158 controls. The mean age of the study group was 34.6 (7.1 SD) years (range 21-42) and 32.1 (7.7 SD) years (range 18-44) for controls. Serum levels of thyroid-stimulating hormone, free thyroxine and the anti-thyroperoxidase and anti-thyroglobulin antibodies were evaluated. RESULTS: Thyroid disorders were identified in 20.9% of the endometriosis group and 26.5% of the control group (P = 0.25). The overall frequency of thyroid dysfunction was 12.2% and 10.8% for the endometriosis and control groups, and the frequency of positive thyroid antibodies, 14.9% and 22.2%, respectively (P = 0.20). Endometriosis stage and infertility history were not associated with thyroid dysfunction and AITD in the study group. CONCLUSIONS: The prevalence of thyroid dysfunction and AITD was similar in the two study groups. Screening for thyroid disturbances in women with endometriosis is not indicated.

Thalidomide is a selective inhibitor of tumor necrosis factor-alpha (TNF-alpha), a cytokine involved in mycobacterial death mechanisms. We investigated the role of this drug in the functional activity of alveolar macrophages in the presence of infection induced by intranasal inoculation of Mycobacterium avium in thalidomide-treated and untreated adult Swiss mice. Sixty animals were inoculated with 5 x 10(6) M. avium by the respiratory route. Thirty animals received daily thalidomide (30 mg/kg mouse) and 30 received water by gavage up to sacrifice. Ten non-inoculated mice were used as a control group. Lots of animals from each group were evaluated until 6 weeks after inoculation. Infection resulted in an increased total number of inflammatory cells as well as increased activity of pulmonary macrophages. Histologically, intranasal inoculation of bacilli resulted in small mononuclear infiltrates located at the periphery of the organ. Culture of lung fragments revealed the presence of bacilli only at the beginning and at the end of the experimental period. Thalidomide administration did not affect the microbiological or histological features of the infection. Thalidomide-treated and untreated animals showed the same amount of M. avium colonies 3 weeks after infection. Although it did not affect bacillary clearance, thalidomide administration resulted in a decreased percent of spread cells and release of hydrogen peroxide, suggesting that factors other than TNF-alpha play a role in the killing of mycobacteria by alveolar macrophages. Thalidomide administration also reduced the number of spread cells among resident macrophages, suggesting a direct effect of the drug on this phenomenon.

BACKGROUND: The study aim was to assess the time elapsed between onset of symptoms and diagnosis of endometriosis, and to identify the factors associated with diagnostic delay in a group of Brazilian women. METHODS: In this retrospective cohort study, 200 women with surgically confirmed endometriosis were interviewed at an endometriosis outpatient clinic. RESULTS: The median (interquartile range) time elapsed from onset of symptoms until diagnosis of endometriosis was 7.0 (range 3.5-12.1) years. The younger the women at onset of symptoms, the longer the period for diagnosis to be made: the median delay was 12.1 (range 8.0-17.2) years in women aged < or =19 years, and 3.3 (range 2.0-5.5) years in women aged > or =30 years. The median time period between onset of symptoms and diagnosis was 4.0 (2.0-6.0) years for women whose main complaint was infertility, but 7.4 (3.6-13.0) years for those with pelvic pain. CONCLUSIONS: The delay in diagnosis of endometriosis was considered to be long, and especially so for young women with pelvic pain. More information relating to endometriosis should be offered to general physicians and gynaecologists in order to reduce the time taken to diagnose this condition.

UNLABELLED: Transseptal left heart catheterization has been performed as an alternative to the retrograde approach since 1958. However, this procedure can result in life-threatening complications, some of which may occur because of insufficient anatomical landmarks. Accordingly, we sought to assess the safety and efficacy of a new transseptal left heart catheterization technique designed for ablation procedures. Specifically, we examined whether electrode catheters could be used as anatomical landmarks, permitting identification of the aortic root and other critical structures. RESULTS: One hundred and eight consecutive patients underwent transseptal left heart catheterization under biplane fluoroscopy during catheter ablation. Electrode catheters positioned in the right atrial appendage, His bundle region, and coronary sinus were used as anatomical landmarks to guide the transseptal unit to the fossa ovalis. The angles of the right anterior and left anterior oblique projections were selected in each patient based on the orientation of the His bundle and coronary sinus catheters. Transseptal left heart catheterization was successfully performed in all patients without complications. In contrast to previous reports, the direction of the needle at the successful puncture site in the last 96 patients varied substantially: 2 o'clock in 13 patients (13 %); 3 o'clock in 43 patients (45 %); and 4 o'clock in 40 patients (42 %). CONCLUSION: The use of electrode catheters as anatomical landmarks and biplane fluoroscopy facilitates transseptal catheterization. This approach can be used safely during catheter ablation procedures.

This study presents the results of T. mentagrophytes inoculation in the cheek pouch of the hamster, an immunologically privileged site. Forty two animals were used: 21 inoculated with 10(6) fungi in the cheek pouch (group 1) and 21 inoculated initially with 10(6) fungi in the foot pad and 15 days later in the cheek pouch, with the same amount of fungi (group 2). Animals were sacrificed at 20 hours, 3, 7, 14, 30, 60, and 120 days; samples from inoculated cheek pouch, and foot pads submitted to the foot pad test (FPT), were collected. Independent of group and time of evolution of infection, animals did not develop delayed hypersensitivity evaluated through the FPT. The pre-inoculation of fungi in the foot pad did not change the morphology of lesions induced in the cheek pouch. Therefore, in animals of group 1 and 2, the introduction of the fungus in the cheek pouch resulted in focal lesion composed of a sterile acute inflammatory infiltrate, with abscess formation that evolved to a macrophagic reaction, and later to resolution even in the absence of immune response detectable by FPT. Our results indicate that in spite of the important role of the immune response in the spontaneous regression of dermatophytosis, other factors are also an integral part in the defense against this fungal infection.

UNLABELLED: Pressure sores are common among bedridden, elderly, or malnourished patients, and may occur in terminal ill patients because of impaired mobility, fecal or urinary incontinence, and decreased healing capacity. The aim of this study was to compare frequency of pressure sores between malnourished and non-malnourished necropsied adults. METHOD: All (n = 201) adults (age >/= 18 years) autopsied between 1986 and 1996 at the Teaching Hospital of Triangulo Mineiro Medical School (Uberaba) were eligible for the study. Gender, race, weight, height and main diagnoses were recorded. Ninety-six cases were excluded because of probable body water retention (congestive heart failure, hepatic insufficiency, nephrotic syndrome) or pressure sores secondary to peripheral vascular ischemia. Body mass index (BMI) was used to define malnourished (BMI < 18.5 kg/m2) and non-malnourished (BMI > 18.5kg/m2) groups. RESULTS: Except for weight (42.5kg; range: 28-57 vs. 60; 36-134.5kg) and BMI (16.9; range: 12.4-18.5 vs. 22.7; range: 18.5-54.6kg/m2), respectively, there were no statistical differences among 43 malnourished and 62 non-malnourished cases in relation to age (54.9 +/- 20.4 vs. 52.9 +/- 17.9 years), percentage of white persons (74.4 vs. 64.5%), male gender (76.7 vs. 69.3%) and main diagnoses. Five malnourished (11. 6%) and 7 (11.5%) non-malnourished cases had pressure sores (p=0.89). CONCLUSION: Pressure sores were equally common findings in necropsied persons with protein-energy malnutrition, as assessed by body mass index.

4'-Hydroxy-7,8-[2-(2-hydroxyisopropyl)dihydrofuran]flavan and 4',7-dihydroxy-8-(3,3-dimethylallyl)flavan, together with 10 known plant constituents, were obtained from the trunk bark of Brosimum acutifolium. Their structures were elucidated by spectroscopic methods, including 2D NMR spectroscopic techniques.

RESUMEN

Pellucidin A, a novel dimeric ArC2 compound, together with dill-apiol have been isolated from the aerial parts of Peperomia pellucida. The structure of pellucidin A was established by 1D and 2D NMR spectroscopy (1H-1H COSY; 1H-13C COSY; DEPT; NOESY and double irradiation) and other spectroscopic techniques. The biogenesis of pellucidin A is also briefly discussed.

The authors investigated the relationship between dermatophytosis and ABO blood groups through blood typing, identification of isolated dermatophytes and specific cellular immune response of 40 individuals carriers of this mycosis. They verified that the fungus Trichophyton rubrum, isolated from 54.5% of the patients, was more frequent in individuals belonging to blood group A. The cellular immune response, evaluated through the trichophytin antigen, was positive in 25% of the studied patients; the presence of immediate reactions (30 minutes) was verified in 35%. The blood group distribution among patients with dermatophytosis and control groups was, respectively: 47.5% X 36% in group A, 40% X 50% in group O, 12. 5% X 11% in group B. Even though the authors have found a higher number of patients belonging to blood group A infected by T. rubrum, these results suggest that there is no statistical evidence that these individuals are more susceptible to dermatophytosis.
