RESUMEN

Population expansion is a global issue, especially for food production. Meanwhile, global climate change is damaging our soils, making it difficult for crops to thrive and lowering both production and quality. Poor nutrition and salinity stress affect plant growth and development. Although the impact of individual plant stresses has been studied for decades, the real stress scenario is more complex due to the exposure to multiple stresses at the same time. Here we investigate using existing evidence and a meta-analysis approach to determine molecular linkages between two contemporaneous abiotic stimuli, phosphate (Pi) deficiency and salinity, on a single plant cell model, the root hairs (RHs), which is the first plant cell exposed to them. Understanding how these two stresses work molecularly in RHs may help us build super-adaptable crops and sustainable agriculture in the face of global climate change.

Harvest date is a critical parameter for producers and consumers regarding agro-industrial performance. It involves a pleiotropic effect controlling the development of other fruit quality traits through finely controlling regulatory mechanisms. Fruit ripening is a process in which various signals and biological events co-occur and are regulated by hormone signaling that produces the accumulation/degradation of multiple compounds. However, the regulatory mechanisms that control the hormone signaling involved in fruit development and ripening are still unclear. To investigate the issue, we used individuals with early, middle and late harvest dates from a peach segregating population to identify regulatory candidate genes controlling fruit quality traits at the harvest stage and validate them in contrasting peach varieties for this trait. We identified 467 and 654 differentially expressed genes for early and late harvest through a transcriptomic approach. In addition, using the Arabidopsis DAP-seq database and network analysis, six transcription factors were selected. Our results suggest significant hormonal balance and cell wall composition/structure differences between early and late harvest samples. Thus, we propose that higher expression levels of the transcription factors HB7, ERF017 and WRKY70 in early harvest individuals would induce the expression of genes associated with the jasmonic acid pathway, photosynthesis and gibberellins inhibition. While on the other hand, the high expression levels of LHY, CDF3 and NAC083 in late harvest individuals would promote the induction of genes associated with abscisic acid biosynthesis, auxins and cell wall remodeling.

RESUMEN Introducción: la estomatitis aftosa recurrente es una enfermedad habitual en las urgencias de Periodontología. Objetivo: evaluar la efectividad de la crema de Calendula officinalis L. al 10%, en el tratamiento de la estomatitis aftosa recurrente, en una clínica estomatológica. Método: se realizó un ensayo no controlado- no aleatorizado, en una población de 15 a 34 años de edad; que acudieron a la consulta de periodoncia de la Clínica Estomatológica Docente "Manuel Angulo Farrán" en Holguín, Cuba, en el período comprendido del año 2015 al 2017. La muestra fue de 100 pacientes, tratada durante 6 días, con consultas cada 48 horas. Resultados: el 59% de los pacientes fueron del sexo femenino, el 63% entre los 25 y 34 años, y en el 97% predominaron las aftas menores. La crema en estudio mejoró los síntomas en el 97% de los pacientes, y el 94% se curó. Conclusiones: la crema de Calendula officinalis L al 10% parece eficaz en el tratamiento de la estomatitis aftosa recurrente, aunque se requieren ensayos clínicos controlados y aleatorizados que lo demuestren de forma definitiva.


ABSTRACT Introduction: recurrent aphthous stomatitis is a frequent disease in periodontology emergencies. Objective: to evaluate the Calendula officinalis L at 10% cream effectiveness, as treatment for recurrent aphthous stomatitis, in a stomatological clinic. Method: non-control, non-randomized trial was conducted in a population between 15 and 34 years old, who attended "Manuel Angulo Farrán" Stomatological Teaching Clinic in Holguín, Cuba, from 2015 to 2017. Sample was 100 patients, treated in consultations every 48 hours, for 6 days. Results: the 59% of the patients were female, 63% between 25 to 34 years, and minor aphthae predominated in the 97%. The studied cream improved symptoms in 97% of patients and 94% were completely cured. Conclusions: Calendula officinalis L at 10% cream seems effective in the treatment of recurrent aphthous stomatitis, although controlled and randomized clinical trials would definitively demonstrate it.

The effect of copper on the expression of genes encoding the ligninolytic enzymes laccase (lcs) and manganese peroxidase (mnp) in Ceriporiopsis subvermispora was evaluated. This metal increased transcript levels of lcs, mnp1 and mnp2. This finding was not unexpected in the case of lcs, since its promoter contains a putative ACE element. Originally characterized in the yeast Saccharomyces cerevisiae, ACE is the target sequence of the ACE1 copper-responsive transcription factor in this microorganism. Analysis of the promoter regions of mnp genes revealed the presence of formerly unnoticed ACE elements. Based on the ace1 gene from Phanerochaete chrysosporium, we isolated and characterized an ACE1-like transcription factor from C. subvermispora (Cs-ACE1) through complementation of a S. cerevisiae ace1Delta strain. Surprisingly, ACE1 factors from both basidiomycetes exhibit substantial differences, not only structurally but also in their ability to complement the aforementioned yeast strain. Specific binding of Cs-ACE1 to its cognate DNA sequence was confirmed by electrophoretic mobility-shift assays.

Se realizó un estudio trasversal en 43 pacientes alcohólicos del Centro Comunitario No 3 de Holguín, en el período de diciembre 2004 a mayo de 2005, con el objetivo de valorar la salud periodontal según la prevalencia de periodontopatías y determinar la asociación entre estas y el tiempo de ingestión de bebidas alcohólicas e higiene bucal. De los centros comunitarios se escogió aleatoriamente el de referencia y la muestra quedó constituida por pacientes alcohólicos desintoxicados ingresados durante ocho semanas en período de deshabituación, quienes fueron incluidos porque dieron su consentimiento. La entrevista y el examen bucal facilitaron el llenado del formulario confeccionado para este propósito. Se utilizaron el índice periodontal de Russell (IP-R) modificado por la Organización Mundial de la Salud y el de análisis de higiene bucal de Love (IAH). A cross-sectional study was carried out in 43 alcoholic patients at number 3 community center in Holguín, from December 2004 to May 2005, aimed at assessing the periodontal behavior, according to the prevalence of periodontal diseases, and to determine the association among these and the time of alcoholic intake and oral hygiene. The sample was constituted by alcoholic patients who were admitted at a hospital during eight weeks. They were detoxified The data were collected through a questionnaire and oral examination. We have previously identified and functionally characterized the transcription factor ACE1 (Pc-ACE1) from Phanerochaete chrysosporium. In Saccharomyces cerevisiae, ACE1 activates the copper-dependent transcription of target genes through a DNA sequence element named ACE. However, the possible target gene(s) of Pc-ACE1 were unknown. An in silico search led to the identification of putative ACE elements in the promoter region of mco1, one of the four clustered genes encoding multicopper oxidases (MCOs) in P. chrysosporium. Since copper exerts an effect at the transcriptional level in MCOs from several organisms, in this work we analysed the effect of copper on transcript levels of the clustered MCO genes from P. chrysosporium, with the exception of the transcriptionally inactive mco3. Copper supplementation of cultures produced an increment in transcripts from genes mco1 and mco2, but not from mco4. Electrophoretic mobility-shift assays revealed that Pc-ACE1 binds specifically to a probe containing one of the putative ACE elements found in the promoter of mco1. In addition, using a cell-free transcription system, we showed that in the presence of cuprous ion, Pc-ACE1 induces activation of the promoter of mco1, but not that of mco2.
